چکیده
Supplementary Methods Plasmids and Constructs pCMXGFP-Dab1, pCMXGFP-Dab1, pCMXGFP-Dab1, pCMXCFP-Dab1, pCMXGFP-Dab1 and pCMXGFP-Dab1 expression plasmids were constructed by cutting out fragments containing the corresponding mutants from pBSK HADab1 (Howell et al., 2000) with EcoRI/XbaI and subcloning into pCMXGFP-p1k1 with EcoRI/NheI. The pCAG-Dab1GFP was described before (Xu et al., 2005). Different phosphorylation site mutants were generated by swapping in BstXI /BglII fragments from the corresponding pCMXGFP-Dab1 mutants. The following constructs were kind gifts from the following individuals: pME18S-Fyn(B) WT and KM(kinase dead), from Tohru Tezuka; pEF-FlagmSOCS1/3, Neil Nathanson; pEF-Flag-mSOCS2, Chris Greenhalgh (Nicholson et al., 1999); pCEFL-hCbl-b-HA and corresponding Cbl, Cbl-3 and myc-E6AP constructs, Stanley Lipkowitz (Ettenberg et al., 1999; Hatakeyama et al., 1997; Keane et al., 1999); pCMX Flag-mLNX, Jane McGlade (Nie et al., 2002); Rnf5, Ze'ev Ronai (Didier et al., 2003); and Hakai, Yasu Fujita (Fujita et al., 2002).

Cullin 5 regulates cortical layering by modulating the speed and duration of Dab1-dependent neuronal migration.
The multilayered mammalian neocortex develops by the coordinated immigration and differentiation of cells that are produced at distant sites. Correct layering requires an extracellular protein, Reelin (Reln), an intracellular signaling molecule, Disabled-1 (Dab1), and an E3 ubiquitin ligase, Cullin-5 (Cul5). Reln activates Dab1, which is then degraded by Cul5. Here we test whether Cul5 regulate...

متن کاملNdel1 Operates in a Common Pathway with LIS1 and Cytoplasmic Dynein to Regulate Cortical Neuronal Positioning
Correct neuronal migration and positioning during cortical development are essential for proper brain function. Mutations of the LIS1 gene result in human lissencephaly (smooth brain), which features misplaced cortical neurons and disarrayed cerebral lamination. However, the mechanism by which LIS1 regulates neuronal migration remains unknown. Using RNA interference (RNAi), we found that the bi...
متن کاملImpaired Reelin-Dab1 Signaling Contributes to Neuronal Migration Deficits of Tuberous Sclerosis Complex.
Tuberous sclerosis complex (TSC) is associated with neurodevelopmental abnormalities, including defects in neuronal migration. However, the alterations in cell signaling mechanisms critical for migration and final positioning of neurons in TSC remain unclear. Our detailed cellular analyses reveal that reduced Tsc2 in newborn neurons causes abnormalities in leading processes of migrating neurons...
متن کاملBDNF Regulates Reelin Expression and Cajal-Retzius Cell Development in the Cerebral Cortex
Cajal-Retzius (CR) cells of the cerebral cortex express receptors for the neurotrophin brain-derived neurotrophic factor (BDNF) and downregulate expression of the extracellular matrix protein Reelin during early postnatal development, coincident with the onset of cortical BDNF expression. During this period, mice lacking BDNF have elevated levels of Reelin in CR cells. Acute BDNF stimulation of...
