چکیده

: BHK-21-cI3 fibroblasts were incubated with [I-3H, U-14C]glucose or 14C]glucose to produce intracellular [3H]NADPH via the phosphogluconate pathway. 3H and 14C were then determined at the th~ee positions of glycerol in glycerol phosphate, saponifiable glycerolipids, alkyl ether glycerolipids and plasmalogens. The 3H/14C ratio at C-2 in glycerol of saponifiable glycerolipids is 2-10 fold greater than in glycerol phosphate and approaches the ratio found in ether-containing glycerolipids. This suggests that a significant fraction of the glycerolipids in BHK-21-cI3 fibroblasts is synthesized via acyl dihydroxyacetone phosphate.

The relative utilization of the acyl dihydroxyacetone phosphate and glycerol phosphate pathways for synthesis of glycerolipids in various tumors and normal tissues.

Rates of phosphatidate synthesis from dihydroxyacetone phosphate via acyl dihydroxyacetone phosphate or glycerol phosphate are compared in homogenates of 13 tissues, most of which are deficient in glycerol phosphate dehydrogenase (EC 1.1.1.8). In all tissues examined, dihydroxyacetone phosphate entered phosphatidate more rapidly via acyl dihydroxyacetone phosphate than via glycerol phosphate. T...

Redundant systems of phosphatidic acid biosynthesis via acylation of glycerol-3-phosphate or dihydroxyacetone phosphate in the yeast Saccharomyces cerevisiae.

In the yeast Saccharomyces cerevisiae lipid particles harbor two acyltransferases, Gat1p and Slc1p, which catalyze subsequent steps of acylation required for the formation of phosphatidic acid. Both enzymes are also components of the endoplasmic reticulum, but this compartment contains additional acyltransferase(s) involved in the biosynthesis of phosphatidic acid (K. Athenstaedt and G. Daum, J...

Mechanisms whereby insulin increases diacylglycerol in BC3H-1 myocytes.

We previously suggested that insulin increases diacylglycerol (DAG) in BC3H-1 myocytes, both by increases in synthesis de novo of phosphatidic acid (PA) and by hydrolysis of non-inositol-containing phospholipids, such as phosphatidylcholine (PC) and phosphatidylethanolamine (PE). We have now evaluated these insulin effects more thoroughly, and several potential mechanisms for their induction. I...

Induction of the peroxisomal glycerolipid-synthesizing enzymes during differentiation of 3T3-L1 adipocytes. Role in triacylglycerol synthesis.

The glycerophosphate backbone for triglyceride synthesis is commonly believed to be created through the conversion of dihydroxyacetone phosphate (DHAP) by glycerophosphate dehydrogenase (GPD) to sn-glycerol 3-phosphate (GP), which is then converted by glycerophosphate acyltransferase (GPAT) to 1-acyl-GP. Consistent with this, GPD and GPAT are highly induced during differentiation of mouse 3T3-L...

Glycerolipid biosynthesis in peroxisomes via the acyl dihydroxyacetone phosphate pathway.

In recent years, acyl dihydroxyacetone phosphate [acyl-DHAPt) has been shown to be a precursor of glycerolipids and glycerol-ether lipids.'.' Acyl-DHAP was discovered as a rapidly labeled lipid that was formed in crude mitochondrial fraction from 3'Pi or y-32P[ATP].3 This rapid labeling was due to the enzymatic dephosphorylation and rephosphorylation of endogenous acyl-DHAP present in the crude...
