Automated hydrolysis of total estrogens in urine from pregnant women.
This method for the hydrolysis of estrogens completes the automated analysis of estrogens in pregnancy urine described by Muir, Ua Conaill, and Ryan [Steroids 13, 719 (1969)]. Pregnancy urines can be processed from the sampling of urine to the recording of fluorescence intensity in 30 mm, at a rate of 20 samples per hour. The presence of glucose does not de crease the apparent hydrolysis products.
متن کاملAn automated method for the determination of estrogens in pregnancy urine.
Ittrich’s method for the determination of total estrogens in urine (1) was adapted for automation on the AutoAnalyzer, using a continuous digester module for the combined operation of phase exchange and color development. The automated method gave good recovery and reproducibility, and correlated well with the manual method. It increased the capacity of this laboratory for estrogen determinatio...
متن کاملUrinary estrogens in pregnancy; improved method for their determination in humans.
NE OF PEE DIFFICULTIES encountered in the determination of urinary estrogens arises from the fact that no procedure so far developed is capable of cleaving the estrogen conjugates quantitatively without destroying any of the free steroids (1). Reporting on a thorough investigation of this subject Van Bruggen (2) stated: “The use of butanol as the solvent and hydrolyzing medium.. . appears to be...
متن کاملA semi-automated fluorometric method for total estrogens in pregnancy urine.
We report a continuous-flow fluorometric method for total urinary estrogens that involves the Kober reaction, with extraction of the reaction product into dichloroethane containing trichloroacetic acid as described by Hahnel and Jones [Clin. Chim. Acta 16, 185 (1967)]. The dichloroethane extraction gives greater stability to the Kober color and sharper separation of aqueous and organic phases. ...
متن کاملOestrogen conjugates of human late-pregnancy urine.
1. The separation of the oestrogen conjugates in late-pregnancy urine into two groups, peaks I and II, by gel filtration on Sephadex G-25 (Beling, 1963) has been shown to be affected by the presence of urate, which delays the elution of peak II conjugates. 2. By reapplication to a Sephadex column, peak I conjugates have been further separated into two groups (peaks IA and IB) and the metabolite...
