چکیده

maintaining proper cholesterol levels. Several recent studies of removal of excess cholesterol in the CNS have focused on the ABC-A1 lipid transporter and its regulation via the LXR nuclear hormone receptor. Cellular cholesterol is hydroxylated to form oxysterols (24hydroxycholesterol in the brain), which bind LXR and promote gene transcription. LXR activation increases levels of ABC-A1 and apoE, which together act to remove cholesterol and other lipids from cells. Recent studies including Liang et al. suggest manipulation of the LXR system alters brain cholesterol homeostasis and apoE levels, and thus could be beneficial in approaches to Alzheimer disease therapeutics.

O-16: Metabolism of Exogenous Fatty Acids, Fatty Acid-Mediated Cholesterol Efflux, PKA and PKC Pathways in Boar Sperm Acrosome Reaction

Background: For understanding the roles of fatty acids on the induction of acrosome reaction which occurs under association of cholesterol efflux and PKA or PKC pathways in boar spermatozoa, metabolic fate of alone and combined radiolabeled 14C-oleic acid and 3H-linoleic acid incorporated in the sperm was compared, and behavior of cholesterol and effects of PKA and PKC inhibitors upon fatty aci...

Cholesterol efflux to apolipoprotein AI involves endocytosis and resecretion in a calcium-dependent pathway.

We previously have described the cAMP-mediated induction of cholesterol and phospholipid efflux from the murine macrophage RAW264 cell line to lipid-free apolipoprotein acceptors. This induction of cholesterol efflux is associated with increased binding and association of apolipoprotein to the cells. In the present study, using primarily apolipoprotein AI (apoAI) as the acceptor, cAMP-dependent...

Deficiency of ABCA1 impairs apolipoprotein E metabolism in brain.

ABCA1 is a cholesterol transporter that is widely expressed throughout the body. Outside the central nervous system (CNS), ABCA1 functions in the biogenesis of high-density lipoprotein (HDL), where it mediates the efflux of cholesterol and phospholipids to apolipoprotein (apo) A-I. Deficiency of ABCA1 results in lack of circulating HDL and greatly reduced levels of apoA-I. ABCA1 is also express...

Regulation of cholesterol homeostasis by the liver X receptors in the central nervous system.

The nuclear oxysterol receptors liver X receptor-alpha [LXRalpha (NR1H3)] and LXRbeta (NR1H2) coordinately regulate genes involved in cholesterol homeostasis. Although both LXR subtypes are expressed in the brain, their roles in this tissue remain largely unexplored. In this report, we show that LXR agonists have marked effects on gene expression in murine brain tissue both in vitro and in vivo...

Purple perilla extracts with α-asarone enhance cholesterol efflux from oxidized LDL-exposed macrophages

The cellular accumulation of cholesterol is critical in the development and progression of atherosclerosis. ATP-binding cassette (ABC) transporters play an essential role in mediating the efflux of excess cholesterol. In the current study, we investigated whether purple Perilla frutescens extracts (PPE) at a non-toxic concentration of 1-10 µg/ml stimulate the induction of the ABC transporters, ...
