چکیده

Puroose Recurrent ocular toxoplasmosis is a major cause of blindness throughout the world. However, the pathogenetic mechanisms of reactivation of latent Toxoplasma gondii (T. gondii) infections are still unknqwn. An important question is whether the recurrent focal inflammatory responses in the retina are primarily directed against the parasite itself or whether autoimmune mechanisms play a role in the reactivations. Methods We designed a study to isolate and characterise the intraocular T cells involved in the recurrent inflammations. We tested a panel of T cell lines and T cell clones (TLCs) that have been derived from the vitreous of 7 patients with active recurrent ocular toxoplasmosis. w While no reactivity was found against retinal antigens, all T cell lines and 52 TLCs generated responded significantly to T. go&ii. The T. gondii-specific TLCs had the CD4+ phenotype, were generally HLA-DR restricted and displayed a heterogeneous T cell receptor usage. Conclusions Our findings suggest that the intraocular cellular immune response in recurrent ocular toxoplasmosis is predominantly T. gondii-specific. These observations are relevant not only to a further understanding of protective immunity and immunopathology but also may have implications for the design of effective vaccines.

Immunologic cross-reactivity in the pathogenesis of ocular onchocerciasis.

PURPOSE Onchocerca volvulus, a filarial worm, is a major cause of infectious blindness and inflammatory eye disease. An autoimmune cause for ocular onchocerciasis has been suggested since the identification of a recombinant antigen of O. volvulus that shows immunologic cross-reactivity with a host ocular component of 44,000 M(r). The aim of this study was to establish the distribution of the cr...

Molecular and immunological characterization of hr44, a human ocular component immunologically cross-reactive with antigen Ov39 of Onchocerca volvulus

Structural similarities between host self-antigens and infectious organisms may be involved in the expression of autoimmune reactivity and development of autoimmune disease. The unique eye pathology associated with Onchocerca volvulus infection, particularly the development of posterior segment lesions, may be promoted by such autoreactive responses. Ov39 is a parasite-derived antigen that has ...

Antigenic mimicry: Onchocerca volvulus antigen-specific T cells and ocular inflammation.

PURPOSE Molecular mimicry has been suggested to play a role in the development of ocular onchocerciasis. The Onchocerca volvulus antigen Ov39 is cross-reactive with the retinal antigen hr44 and induces ocular inflammation in rats after immunization. This study was undertaken to determine whether Ov39-derived T-cell lines, which proliferate in response to stimulation with hr44, can transfer dise...

IgE antibodies in human onchocerciasis. Application of a newly developed radioallergosorbent test (RAST).

A radioallergosorbent test (RAST) was developed to detect IgE antibodies against adult Onchocerca volvulus antigens coupled to CnBr-activated Sepharose. Twenty-four out of 25 (96%) onchocerciasis sera were reactive. The lower limit of sensitivity was estimated to be at approx. 3 ng/ml IgE antibodies. Tests of sera from patients with non-filarial helminth infections showed much less cross-reacti...

Humoral autoimmune response against S-antigen and IRBP in ocular onchocerciasis.

Autoimmune mechanisms are thought to play a role in the pathogenesis of the chorioretinal changes in ocular onchocerciasis. In this study, the involvement of autoimmunity against retinal antigens in developing chorioretinitis was investigated. Serum levels of autoantibodies, directed against human S-antigen and interphotoreceptor retinoid-binding protein (IRBP), were determined in patients with...
