Comparison of antifibrillatory potency of certain antimalarial drugs with quinidine and procaine amide.

چکیده
The procedure used to assay the potency of antifibrillatory drugs has been described by DiPalma and colleagues. A brief summary will suffice for this report. Cats weighing between 2 and 3 Kg. were anesthetized with intraperitoneal Dial (Ciba). Artificial respiration was maintained at an even level with a suitable pump, and the heart was exposed and suspended in :i pericardinl cradle. Clip electrodes were attached to the right atrium. A current of 600 impulses per minute was supplied by a thyratron stimulator. The current was conducted through a calibrated and variable resistance unit which permitted the determination of the liminal amount of current which would just induce atrial fibrillation. On the average this varied between 2 and S milliamperes. In a typical assay the control fibrillation threshold

or quinidine. Two dose schedules were employed; procaine amide, 3.0 and 6.0 g daily and quinidine, 1.2 and 1.8 g daily. Though adequate and even high drug blood levels were reached, an effective antiarrhythmic response was observed in only eight patients receiving procaine amide and in seven of those taking quinidine. These modest successful results were associated with a high incidence of trou...
متن کاملCentrally mediated effects of cardiac drugs: strophanthin-K, quinidine and procaine amide.
The extra-cardiac sites of action of strophanthin-K, quinidine and procaine amide have been investigated by injection of varying doses into the third ventricle of unanesthetized dogs. A profuse autonomic nervous system discharge was produced. Strophanthin-K produced a paroxysmal ventricular tachycardia resulting in death by ventricular fibrillation. Such effects could be blocked by prior intrav...
متن کاملThe action of procaine and procaine amide on the heart.
The depressant action of procaine on the heart muscle has been the subject of several clinical and experimental studies (Wiggers and Wegria, 1940; Burstein, 1946; Uhley and Wilburne, 1948; Schaffer et al., 1950). Procaine was found useful in the treatment of cardiac arrhythmias occurring in anesthetized patients during cardiac surgery. However, its usefulness in the management of clinical arrhy...
متن کاملThe effects of intravenous procaine and procaine amide (pronestyl) upon ectopic ventricular tachycardia accompanying acute myocardial infarction.
In tests using dogs with ectopic ventricular tachycardia resulting from myocardial infarction, procaine has been found to be without practical value as an ectopic impulse suppressor agent. Procaine amide has proved to be very effective in reducing the ectopic rate of high frequency tachycardias to low levels, thus eliminating danger of death from the tachycardia. Procaine amide usually does not...
متن کاملProcaine amide therapy in auricular flutter with 1:1 A-V conduction.
Procaine amide hydrochloride was first introduced clinically by Mark et al. (1950). They drew attention to its effectiveness in the control of ventricular arrhythmias, especially paroxysmal ventricular tachycardia. This has since been amply confirmed. It is the drug of choice in the treatment of paroxysmal ventricular tachycardia, taking precedence over the more toxic and unpredictable quinidin...
