Corrigendum: An apoptosis-enhancing drug overcomes platinum resistance in a tumour-initiating subpopulation of ovarian cancer

چکیده
After reanalysing the RNA sequencing data presented in Supplementary Data 3 of this Article, we have discovered that 341,056 of the 730,620 expression values were inadvertently paired with incorrect gene names. These incorrect values were used in Figure 3c–e and Supplementary Tables 4,5,7 and 8 and led us to conclude that there are differences in the expression of DNA repair and apoptosis genes between CA125-negative and CA125-positive cells. We have now reanalysed the corrected data, and present a revised version of Figure 3 below. We find that, of the DNA repair and apoptosis genes reported in Figure 3, none display a statistically significant difference in expression between CA125-negative and CA125-positive cells at the transcript level. However, at the protein level, cIAPs, the target proteins for Birinapant, are nevertheless expressed at substantially higher levels by CA125-negative cells, implying that the regulatory mechanisms of cIAP protein expression may be different from transcriptional regulation in CA125-negative cells. Therefore, the overall conclusion in regard to therapy, that birinapant strongly sensitizes CA125-negative cells to carboplatin-induced death due to their high cIAP protein expression, remains unaffected by the error in RNA-sequencing data analysis. Corrected versions of Supplementary Data 3 and Supplementary Tables 4,5,7 and 8 are available from the HTML version of this Corrigendum. DOI: 10.1038/ncomms10703 OPEN

An apoptosis-enhancing drug overcomes platinum resistance in a tumour-initiating subpopulation of ovarian cancer
High-grade serous ovarian cancers (HGSCs) are deadly malignancies that relapse despite carboplatin chemotherapy. Here we show that 16 independent primary HGSC samples contain a CA125-negative population enriched for carboplatin-resistant cancer initiating cells. Transcriptome analysis reveals upregulation of homologous recombination DNA repair and anti-apoptotic signals in this population. Whil...

Ovarian cancer is the most lethal gynecological cancer in which cisplatin-based treatment plays fundamental role as the first line chemotherapy option. However, development of platinum-resistance is a critical and poorly understood problem in ovarian cancer treatment. Although in vitro generation of platinum-resistant ovarian cancer cell lines is a long established approach to uncover the molec...

Paclitaxel is a current standard chemotherapeutic drug for ovarian cancer with several side effects. Recurrences of drug resistant clones have been considered the serious problem in the failure of chemotherapy. Medicinal marine natural products have been intensively proposed as diverse chemotherapeutic agents. Therefore there is an affinity to find efficient modality to overwhelm ovarian cancer...

BACKGROUND Ovarian cancer is the leading cause of death for gynecological cancers and the 6th cause of women cancer death in developed countries. The late stage detection, the peritoneal dissemination and the acquisition of resistance against carboplatin are the main reasons to explain this poor prognosis and strengthen the need of alternative treatments to improve the management of ovarian can...

The development of drug resistance remains one of the major hurdles in cancer chemotherapy, particularly so for ovarian cancer. Combination of drugs acting synergistically in combination can offer a means of overcoming drug resistance. In this study, two tumour-active phytochemicals, quercetin and thymoquinone, were combined with two platinum drugs, cisplatin and oxaliplatin, with the aim of pr...
