چکیده

Here I recap the scientific and personal background of the delineation of the amyloid cascade hypothesis for Alzheimer's disease that I wrote with Gerry Higgins and the events leading to the writing of that influential review.

P135: The Role of Amyloid Beta-Peptides and Tau Protein in Alzheimer\'s Disease

Alzheimer's desease is the most common age-related neurodegenerative disorder, and cognitive problems such as defects in learning and memory are of its symptoms.  Among the factors involved in the pathogenesis of the disease are biochemical disorders in protein production, oxidative stress, decreased acetylcholine secretion and inflammation of the brain tissue. Extra-neuronal accumulation ...

Fleshing out the amyloid cascade hypothesis: the molecular biology of Alzheimer's disease

Alzheimer's disease (AD) is a disorder of two pathologies- plaques and tangles. The former have as a key constituent amyloid protein and the latter the microtubule-associaied protein tau. Genetics has demonstrated that changes in either protein are sufficient to cause dementia. The amyloid cascade hypothesis proposes that plaque-related changes precede tangle-related changes and positions amylo...

The Effect of Endurance Training on the Expression of PRDX6 and KAT2B Genes in Hippocampus of Beta Amyloid-Induced Rat Model of Alzheimer's Disease: An Experimental Study

Background and Objectives: Alzheimer's disease is the most common form of dementia. KAT2B (Lysine Acetyltransferase 2B) is a mitochondrial protein known as mitochondria clearing control organ by mitophagy. PRDX6 (Peroxiredoxin 6) is a key regulator of mitophagy and plays a critical role in maintaining mitochondrial ROS (Reactive oxygen species) homeostasis. Therefore, the purpose of this study ...

Alzheimer's Disease and the Amyloid Cascade Hypothesis: A Critical Review

Since 1992, the amyloid cascade hypothesis has played the prominent role in explaining the etiology and pathogenesis of Alzheimer's disease (AD). It proposes that the deposition of β-amyloid (Aβ) is the initial pathological event in AD leading to the formation of senile plaques (SPs) and then to neurofibrillary tangles (NFTs), neuronal cell death, and ultimately dementia. While there is substan...

A "mitochondrial cascade hypothesis" for sporadic Alzheimer's disease.

Alzheimer's disease (AD) includes etiologically heterogeneous disorders characterized by senile or presenile dementia, extracellular amyloid protein aggregations containing an insoluble amyloid precursor protein derivative, and intracytoplasmic tau protein aggregations. Recent studies also show excess neuronal aneuploidy, programmed cell death (PCD), and mitochondrial dysfunction. The leading A...
