Feasibility study on the use of polyallyldiglycol-carbonate cell dishes in TUNEL assay for alpha particle radiobiological experiments

چکیده
In the present work, we have studied the feasibility of a method based on polyallyldiglycol-carbonate (PADC) films to investigate the effects of alpha particles on HeLa cervix cancer cells. Thin PADC films with thickness of about 20 lm were prepared from commercially available CR-39 films by chemical etching to fabricate custom-made petri dishes for cell culture, which could accurately record alpha particle hit positions. A special method involving ‘‘base tracks’’ for aligning the images of cell nuclei and alpha particle hits has been proposed, so that alpha particle transversals of cell nuclei can be visually counted. Radiobiological experiments were carried out to induce DNA damages, with the TdT-mediated dUTP Nick-End Labeling (TUNEL) fluorescence method employed to detect DNA strand breaks. The staining results were investigated by flow cytometer. The preliminary results showed that more strand breaks occurred in cells hit by alpha particles with lower energies. Moreover, large TUNEL positive signals were obtained even with small percentages of cells irradiated and TUNEL signals were also obtained from non-targeted cells. These provided evidence for the bystander effect. 2007 Elsevier B.V. All rights reserved.

Alpha-particle fluence in radiobiological experiments
Two methods were proposed for determining alpha-particle fluence for radiobiological experiments. The first involved calculating the probabilities of hitting the target for alpha particles emitted from a source through Monte Carlo simulations, which when multiplied by the activity of the source gave the fluence at the target. The second relied on the number of chemically etched alpha-particle t...
متن کاملStudy of influence of catechins on bystander responses in alpha-particle radiobiological experiments using thin PADC films
In this study, Chinese hamster ovary (CHO) cells were cultured in custom-made petri dishes with thin PADC films as substrates. Alpha particles with energies of 5 MeV were then irradiated from the bottom of PADC films. The DNA strand breaks in the bystander cells induced by irradiation were quantified with the use of terminal dUTP transferase-mediated nick end-labeling (TUNEL) assay. To study th...

A brief review on the progress on in vivo studies of α-particle radiation effects using zebrafish embryos was given in this chapter. The zebrafish, Danio rerio, a small vertebrate from Southeast Asia, has become a preferred model for studying human disease. The main challenges of these α-particle radiobiological experiments included quantification of alpha-particle dose. In particular, speciall...

One approach for a particle radiobiological experiments is to use solid-state nuclear track detectors (SSNTDs) as support substrates to record the a particle hit positions on the targets. To facilitate accurate characterization of the hit positions as well as the incident a particle energies, micro-collimators are required in these experiments to restrict the incidentaparticles to thosewith sma...
متن کاملSimple preparation of thin CR-39 detectors for alpha-particle radiobiological experiments
Alpha-particle radiobiological experiments involve irradiating cells with alpha particles and require accurate positions where the alpha particles hit the cells. In the present work, we prepared thin CR-39 detectors from commercially available CR-39 SSNTDs with a thickness of 100 lm by etching them in 1 N NaOH/ethanol at 40 C to below 20 lm. The desired final thickness was achieved within 8 h. ...
