چکیده

pUO-StVR2 is a derivative of pSLT, the virulence plasmid specific of Salmonella enterica serovar Typhimurium, which confers multidrug resistance. This plasmid is widespread among closely related isolates of S. Typhimurium, and often coexists with other plasmids like pStR12. The latter belongs to incompatibility group IncI1, was assigned to ST48 by pMLST (plasmid multilocus sequence typing), and confers resistance to streptomycin/spectinomycin, chloramphenicol, trimethoprim and sulphonamides, with the responsible genes (aadA1/aadA2, cmlA1, dfrA12 and sul3) located on a sul3-class 1 integron. When using clinical isolates of S. Typhimurium containing one (pUO-StVR2) or both (pUO-StVR2 and pStR12) plasmids as donors and Escherichia coli K12 J53 resistant to rifampicin as recipient, the conjugation frequencies of pUO-StVR2 and pStR12 were 10⁻⁸ and 10⁻³-10⁻⁵ transconjugants/donor, respectively, while the transfer frequency of pUO-StVR2 increased 10² up to 10⁵ times through mobilization by pStR12, depending on the donor strain and experimental conditions. Mobilization of pUO-StVR2, a plasmid which encodes virulence and resistance functions, by compatible plasmids which coexist in the same bacterium can facilitate the spread of these properties in S. Typhimurium, one of the most common serovars of S. enterica.

Characterization of pUO-StVR2, a virulence-resistance plasmid evolved from the pSLT virulence plasmid of Salmonella enterica serovar Typhimurium.

pUO-StVR2 is a virulence-resistance plasmid which originated from pSLT of Salmonella enterica serovar Typhimurium through acquisition of a complex resistance island, flanked by regions that provide a toxin-antitoxin system and an iron uptake system. The presence of resistance and virulence determinants on the same plasmid allows coselection of both properties, potentially increasing health risks.

Bile-induced curing of the virulence plasmid in Salmonella enterica serovar Typhimurium.

Exposure to bile induces curing of the virulence plasmid in Salmonella enterica serovar Typhimurium (pSLT). Disruption of the ccdB gene increases pSLT curing, both spontaneous and induced by bile, suggesting that the pSLT ccdAB genes may encode a homolog of the CcdAB addiction module previously described in the F sex factor. Unlike the F sex factor, synthesis of pSLT-encoded pili does not confe...

Complete Nucleotide Sequences of Virulence-Resistance Plasmids Carried by Emerging Multidrug-Resistant Salmonella enterica Serovar Typhimurium Isolated from Cattle in Hokkaido, Japan

In the present study, we have shown that virulence-resistance plasmids from emerging multidrug-resistant isolates of Salmonella enterica serovar Typhimurium were derived from a virulence-associated plasmid, essential for systematic invasiveness of S. Typhimurium in mice (pSLT), through acquisition of a large insert containing a resistance island flanked by IS1294 elements. A bla CMY-2-carrying ...

Diversity of Plasmids Encoding Virulence and Resistance Functions in Salmonella enterica subsp. enterica Serovar Typhimurium Monophasic Variant 4,[5],12:i:- Strains Circulating in Europe

Plasmids encoding resistance and virulence properties in multidrug resistant (MDR) Salmonella enterica (S.) serovar Typhimurium monophasic variant 4,[5],12:i:- isolates recovered from pigs and humans (2006-2008) in Europe were characterised. The isolates were selected based on the detection by PCR-amplification of S. Typhimurium virulence plasmid pSLT genes and were analysed by multi-locus sequ...

Stabilization of the Virulence Plasmid pSLT of Salmonella Typhimurium by Three Maintenance Systems and Its Evaluation by Using a New Stability Test

Certain Salmonella enterica serovars belonging to subspecies I carry low-copy-number virulence plasmids of variable size (50-90 kb). All of these plasmids share the spv operon, which is important for systemic infection. Virulence plasmids are present at low copy numbers. Few copies reduce metabolic burden but suppose a risk of plasmid loss during bacterial division. This drawback is counterbala...
