چکیده

On the basis of this plot it was found (1) that, with a preparation of hog pancreas as the source of esterase, an apparent deviation from theoretical behavior was the result of the simultaneous action of two enzymes on the substrate. In the present investigation it is found that the curves obtained with horse liver esterase display two rather sharp discontinuities that cannot be ascribed to simultaneous action of two or more enzymes. Evidence is presented that one of these discontinuities is caused by a change in the substrate and the other by a change in the kinetic constants of a single enzyme system at a definite (critical) substrate concentration.

Relationships between Substrate Promiscuity and Chiral Selectivity of Esterases from Phylogenetically and Environmentally Diverse Microorganisms

Substrate specificity and selectivity of a biocatalyst are determined by the protein sequence and structure of its active site. Finding versatile biocatalysts acting against multiple substrates while at the same time being chiral selective is of interest for the pharmaceutical and chemical industry. However, the relationships between these two properties in natural microbial enzymes remain unde...

The Role of Highly Conserved Tryptophan in the Sixth Conserved Region at Substrate Specificity of α- amylase

Early in this study, an α-Amylase from Bacillus megaterium WHO (BMW) was isolated from hot springs of Ramsar (North of Iran), and its gene was cloned in E.coli. Based on its conserved sequence regions and substrate specificity, it was classified as intermediary group enzymes with the specificity of oligo-1,6-glucosidase and neopullulanase subfamilies. In the sixth conserved re...

Two novel deep-sea sediment metagenome-derived esterases: residue 199 is the determinant of substrate specificity and preference

BACKGROUND The deep-sea environment harbors a vast pool of novel enzymes. Owing to the limitations of cultivation, cultivation-independent has become an effective method for mining novel enzymes from the environment. Based on a deep-sea sediment metagenomics library, lipolytic-positive clones were obtained by activity-based screening methods. RESULTS Two novel esterases, DMWf18-543 and DMWf18...

Characterization of Esterases of Tamarindus indica Seeds

Germinating seeds of Tamarindus indica synthesizes various enzymes which are required for the degradation of seed reserves such as xyloglucans, fatty acid esters and proteins. Among these, esterases, belonging to a group of hydrolytic enzymes catalyze the hydrolysis of various types of esters. They play an important role in cell expansion as well as detoxification of xenobiotics and many agroch...

The Rote of Plant and Microbial Hydrolytic Enzymes in Pesticide Metabolism

esters with carbonyl, phosphoryl, and thionyl linkages, are subject to enzymatic hydrolysis. Pesticide hydrolysis by esterases and amidases from plants and microorganlsms can serve as a deloxification or activation mechanism that can govern pesticide sel€ctivity or resistance, and initiate or determine the rate of pesticide biodegradation in the environment. Substrate specificity of esterases a...
