h-Barrel membrane protein folding and structure viewed through the lens of a-hemolysin

چکیده
The h-barrel is a transmembrane structural motif commonly encountered in bacterial outer membrane proteins and pore-forming toxins (PFTs). a-Hemolysin (aHL) is a cytotoxin secreted by Staphylococcus aureus that assembles from a water-soluble monomer to form a membrane-bound heptameric h-barrel on the surface of susceptible cells, perforating the cell membranes, leading to cell death and lysis. The mechanism of heptamer assembly, which has been studied extensively, occurs in a stepwise manner, and the structures of the initial, monomeric form and final, membrane-embedded pore are known. The toxin’s ability to assemble from an aqueous, hydrophilic species to a membrane-inserted oligomer is of interest in understanding the assembly of PFTs in particular and the folding and structure of h-barrel membrane proteins in general. Here we review the structures of the monomeric and heptamer states of LukF and aHL, respectively, the mechanism of toxin assembly, and the relationships between aHL and nontoxin h-barrel membrane proteins. Computer Aided Molecular Modeling Of Membrane Metalloprotease
Molecular modeling is a set of computational techniques for construction of 3D structure of a protein especially membrane bound proteins whose structures can not be elucidated using experimental techniques. These techniques has been applied in the study of membrane metalloproteases for comparing wild and mutated enzymes, docking inhibitors in the catalytic site and examination of binding pocket...

Figure 1: Looking down through the pore of Alpha-hemolysin. Alpha-hemolysin, pdb designation 7ahl, is a heptameric transmembrane pore. Being a transmembrane protein it is fairly large, approximately 100 Å in size. The main feature of the protein is the beta-barrelFigure 1. This protein is currently being manipulated in DNA nanopore research. The beta-barrel has an opening of about 26 Å. The sig...
متن کاملThe transition state for folding of an outer membrane protein.
Inspired by the seminal work of Anfinsen, investigations of the folding of small water-soluble proteins have culminated in detailed insights into how these molecules attain and stabilize their native folds. In contrast, despite their overwhelming importance in biology, progress in understanding the folding and stability of membrane proteins remains relatively limited. Here we use mutational ana...

The effects of sodium n-dodecyl sulphate (SDS) on the structure of histone HI has been studied by a combination of e:quilibrium dialysis, U.V. spectroscopy ; polyacrylamide gel electrophoresis, protein titration and viscometery techniques using, 2.5 mM phosphate buffer, pH 6.4. The interaction of H, and SDS in contrast tomanyothel-protein-SDS interactions is organized between V 40 to 70. A...
متن کاملSecondary and tertiary structure formation of the beta-barrel membrane protein OmpA is synchronized and depends on membrane thickness.
The mechanism of membrane insertion and folding of a beta-barrel membrane protein has been studied using the outer membrane protein A (OmpA) as an example. OmpA forms an eight-stranded beta-barrel that functions as a structural protein and perhaps as an ion channel in the outer membrane of Escherichia coli. OmpA folds spontaneously from a urea-denatured state into lipid bilayers of small unilam...
