Differentiating between GLP-1 receptor agonists and DPP-4 inhibitors

چکیده

Article points 1. Dipeptidyl peptidase-4 inhibitors (DPP-4Is) inhibit the activity of DPP-4 to prolong the effects of native glucagon-like peptide-1 (GLP-1), whereas GLP-1 receptor agonists (GLP-1RAs) provide activation of the GLP-1 receptor while remaining resistant to degradation by DPP-4. 2. Both DPP-4Is and GLP-1RAs provide glycaemic control in people with type 2 diabetes, and GLP-1RAs are also associated with significant weight loss. 3. Differences between DPP-4Is and GLP-1RAs include efficacy, tolerability, mechanism of action, and administration route. Reductions in HbA1c and weight are generally greater with GLP-1RAs.

Differentiating among incretin-based therapies in the management of patients with type 2 diabetes mellitus

The glucagon-like peptide-1 receptor (GLP-1R) agonists and dipeptidyl peptidase-4 (DPP-4) inhibitors have become important options for the management of patients with type 2 diabetes mellitus. While the GLP-1R agonists and DPP-4 inhibitors act on the incretin system to regulate glucose homeostasis, there are important clinical differences among the five agents currently available in the U.S. Fo...

Pathogenesis and management of postprandial hyperglycemia: role of incretin-based therapies

Postprandial plasma glucose concentrations are an important contributor to glycemic control. There is evidence suggesting that postprandial hyperglycemia may be an independent risk factor for cardiovascular disease. Glucagon-like peptide-1 (GLP-1) receptor agonists and dipeptidyl peptidase-4 (DPP-4) inhibitors are antidiabetic agents that predominantly reduce postprandial plasma glucose levels....

GLP-1 receptor agonists vs. DPP-4 inhibitors for type 2 diabetes: is one approach more successful or preferable than the other?

BACKGROUND In patients with type 2 diabetes (T2D), incretin-based therapies improve glycaemic control with low incidence of hypoglycaemia and without weight gain, both advantages over traditional add-ons to metformin. Dipeptidyl peptidase-4 (DPP-4) inhibitors are administered orally and provide a physiological increase in glucagon-like peptide-1 (GLP-1) levels, while GLP-1 receptor agonists (GL...

Cardiovascular actions of incretin-based therapies.

Glucagon-like peptide-1 receptor (GLP-1R) agonists and dipeptidyl peptidase-4 (DPP-4) inhibitors represent 2 distinct classes of incretin-based therapies used for the treatment of type 2 diabetes mellitus. Activation of GLP-1R signaling or inhibition of DPP-4 activity produces a broad range of overlapping and unique cardiovascular actions. Native GLP-1 regulates cardiovascular biology via activ...

Dipeptidylpeptidase-4 (DPP-4) inhibitors are favourable to glucagon-like peptide-1 (GLP-1) receptor agonists: yes.

The pharmacological treatment of type 2 diabetes (T2DM) is becoming increasingly complex, especially since the availability of incretin-based therapies. Compared with other glucose-lowering strategies, these novel drugs offer some advantages such as an absence of weight gain and a negligible risk of hypoglycaemia and, possibly, better cardiovascular and β-cell protection. The physician has now ...
