چکیده

Supercritical Fluid Chromatography is an alternative and complementary method to HPLC for the determination of impurities in pharmaceutical products. This study evaluated the performance of the Agilent 1260 Infinity Analytical SFC system for the analysis of thiourea in two model compounds. The study included both isocratic and modifier gradient modes. The resulting data showed excellent linearity and repeatability. Retention time repeatability was better than 0.1%. Limits of detection for the impurity determination were below 0.01% w/w relative to the main compound, and the RSDs of peak area were below 1% at the 0.5% concentration level and below 5% at the 0.05% concentration level in all cases. These values are well within acceptable method validation ranges. Authors Melissa N. Feed Speed and Overfeed Volume New Parameters for Injection in Supercritical Fluid Chromatography

This Technical Overview demonstrates and discusses the influence of the injection parameters Feed Speed and Overfeed Volume on chromatographic separation. These parameters were introduced with the Agilent 1260 Infinity II Supercritical Fluid Chromatography (SFC) Multisampler. The influence on the isocratic separation of closely eluting compounds, shown for different injection volumes commonly u...

Superior sensitivity by seamlessly interfacing to the Agilent 6100

The applicability of Supercritical Fluid Chromatography (SFC) can be substantially enhanced by coupling SFC to Mass Spectrometric detection (MSD). In this Technical Overview, two instrument configurations are described that can be used to connect the Agilent 1260 Infinity Analytical SFC to a mass spectrometer either using an atmospheric pressure chemical ionization (APCI) or an electrospray (ES...

Agilent Application Solution Transfer of a USP method for prednisolone from normal phase HPLC to SFC using the Agilent 1260 Infinity Hybrid SFC/UHPLC System

Normal phase liquid chromatography (LC) methods often have long run times and involve environmentally toxic/costly solvents. Supercritical fluid chromatography (SFC) methods on the other hand are faster, inexpensive, and eco-friendly. SFC involves the use of low viscosity supercritical carbon dioxide that can be operated at flow rates up to 3x higher than LC without losing separation efficiency...
