Precipitin Tests with Phosphatide Fractions of Tubercle Bacilli *

چکیده
Many types of precipitin test for tuberculosis based on the use of tuberculin and various extracts have been developed by previous investigators. In general these tests have been unsatisfactory because of their lack of specificity. In 1929 Doan' reported a precipitin test for tuberculosis in which the phosphatide fraction isolated from the tubercle bacillus by Anderson2 was used. His results indicated that the test was of considerable diagnostic value. Employing essentially the method developed by Doan3, we have made a series of tests on material obtained from 233 patients. In this study, tests were made with phosphatide antigenic dilutions only; no antiphosphatide serum was used. The phosphatide fractions used were kindly supplied by the H. K. Mulford Company and were listed by them as follows:

The Proteins in Unheated Culture Filtrates of Human Tubercle Bacilli
1. Only two serologically different proteins were found in the unheated culture filtrates of both virulent and slightly virulent tubercle bacilli. One of them was the protein which had a sedimentation constant of 3.4 S, and the other was in filtrate fractions with a constant of 2 S. 2. That these proteins were distinct was demonstrated by three methods: quantitative precipitin and precipitin ab...

1. The clasmatocyte, the cell with the power of fragmenting tubercle bacilli, the cell making the lesion of the so called interstitial pneumonia, has been shown to be the overwhelming response to the special protein fractions, 304 and 903. Multiple hemorrhages, high fever, and toxicity have marked the use of these fractions in every instance. 2. The epithelioid and the giant cell of the Langhan...
متن کاملEnzymes as Factors in Resistance to Tuberculosis
Jobling and Petersen10 presented evidence as long ago as 1914 that the lipoids of tubercle bacilli inhibit proteolytic enzymes. More recently, Peck"5 has shown that these lipoids also inhibit crystallized trypsin and that the higher the molecular weight of the lipoid the greater is its interference with the enzyme function. Further investigation involving fractions of tubercle bacilli and enzym...
متن کاملThe Effect of Some of the Chemical Constituents of Tubercle Bacilli on the Protoplasm of Amoeba Dubia
1. Protein fractions derived from tubercle bacilli are toxic to the interior of Amoeba dubia but have no action on the plasmalemma. 2. Phosphatide fractions dissolve the plasmalemma but have no effect on the internal cytoplasm. 3. The fatty acid fraction has a marked solvent action on the plasmalemma when brought in contact with the surface of the cell. When injected it may slowly penetrate the...
متن کاملPrecipitins for the Tuberculin Proteins of Acid-fast Bacteria.
Seibert (1930) found that the tuberculin proteins of the three varieties of tubercle bacillus could be distinguished by the precipitin test. If this method could be adapted for identifying tubercle bacilli, it would be preferable to animal inoculation. The behavior of the tuberculin proteins of acid-fast bacteria in the precipitin test is interesting not only as a possible means of identificati...
