چکیده

Expressions of the genes involved in the cytokinin (CK) phosphorelay signaling circuitry in petunia (Petunia hybrida Vilm.) were investigated with regard to the large-flowered trait induced by the semidominant Grandiflora (G) gene. We isolated three cDNAs encoding type-A response regulators (RRs, PhRR1-3) and one cDNA encoding CK receptor histidine protein kinase (PhHK). Expression levels of PhRR1-3 and PhHK in the developing corolla were markedly higher in large-flowered cultivars with the Gg genotype than in smalland medium-flowered cultivars with the gg genotype. Higher expression levels of PhRR1-3 and PhHK in the Gg than in the gg genotypes was also consistent in BC4 plants in which the genetic background except for G allele was mostly uniform. These results suggest that Gg genotype induces a high expression level of PhRR1-3 and PhHK. These changes in expression caused by the G allele were the same as those caused by the CK response. Since type-A RRs repress CK signaling, the G-allele-induced high expression levels of PhRR1-3 presumably have partial inhibitory effects on G-alleleinduced corolla enlargement.

Comparison of Benzyl Adenine Metabolism in Two Petunia hybrida Lines Differing in Shoot Organogenesis.

The uptake and metabolism of the cytokinin benzyl adenine (BA) was compared in two lines of Petunia hybrida Vilm. differing in their shoot organogenic response. Leaf transfer experiments using shoot induction medium containing 4.4 micromolar BA showed that leaf explants from petunia line St40 required a shoot induction period of 6 to 10 days for commitment to shoot organogenesis; whereas leaf e...

A natural antisense transcript of the Petunia hybrida Sho gene suggests a role for an antisense mechanism in cytokinin regulation

The Sho gene from Petunia hybrida encodes an enzyme responsible for the synthesis of plant cytokinins. The 3' region of the Sho gene contains a promoter in the opposite orientation that produces a partially overlapping antisense transcript. Although Sho expression varies significantly in individual cell types, the sense and antisense transcript levels maintain a stable ratio in most tissue type...

Overproduction of cytokinins in petunia flowers transformed with P(SAG12)-IPT delays corolla senescence and decreases sensitivity to ethylene.

Plant senescence is regulated by a coordinated genetic program mediated in part by changes in ethylene, abscisic acid (ABA), and cytokinin content. Transgenic plants with delayed senescence are useful for studying interactions between these signaling mechanisms. Expression of ipt, a cytokinin biosynthetic gene from Agrobacterium tumefaciens, under the control of the promoter from a senescence-a...

The role of ABCG-type ABC transporters in phytohormone transport

Plant hormones (phytohormones) integrate endogenous and exogenous signals thus synchronizing plant growth with environmental and developmental changes. Similar to animals, phytohormones have distinct source and target tissues, hence controlled transport and focused targeting are required for their functions. Many evidences accumulated in the last years about the regulation of long-distance and ...

Functional Characterization of Duplicated SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1-Like Genes in Petunia

Flowering time is strictly controlled by a combination of internal and external signals that match seed set with favorable environmental conditions. In the model plant species Arabidopsis thaliana (Brassicaceae), many of the genes underlying development and evolution of flowering have been discovered. However, much remains unknown about how conserved the flowering gene networks are in plants wi...
