چکیده
Pierce, William A., Jr. (Tulane University, New Orleans, La.). Electrophoretic separation of constituents of partially purified M protein of Streptococcus pyogenes. J. Bacteriol. 88:912-921. 1964.-Partially purified M protein of a group A, type 12 strain of Streptococcus pyogenes was studied by use of chemical, electrophoretic, and immunological techniques. It was demonstrated in immunodiffusion tests that the antigen contains multiple precipitating components. The type-specific antigen was identified, and evidence was presented that, in some instances at least, the cross-reactions observed between this type 12 M protein and heterologous antisera in immunodiffusion tests involve contaminating antigens rather than the component which precipitates with adsorbed homologous-typing antiserum. In passive hemagglutination tests where M protein was adsorbed to tanned sheep erythrocytes, it was found that antisera suitably adsorbed to show good specificity in capillary precipitin tests nevertheless still contain cross-reactive antibodies which are detectable by this more sensitive technique. Electrophoresis on starch paste separates some of the components of partially purified M protein, so that a fraction can be obtained which has fewer precipitating antigens, as determined in immunodiffusion tests, and which is less cross-reactive in passive hemagglutination tests with heterologous unadsorbed antistreptococcal antisera.

Expression of Recombinant Streptokinase from Streptococcus Pyogenes and Its Reaction with Infected Human and Murine Sera
Objective(s): Streptokinase (SKa) is an antigenic protein which is secreted by Streptococcus pyogenes. Streptokinase induces inflammation by complement activation, which may play a role in post infectious diseases. In the present study, recombinant streptokinase from S. pyogenes was produced and showed that recombinant SKa protein was recognized by infected human sera using Western blot analy...
متن کاملCloning and expression of the enzymatic region of Streptococcal hyaluronidase
Objective(s): Streptococcus pyogenes produces extracellular hyaluronidase enzyme. This enzyme is directly associated with the spread of the organism during infection. The objective of the present study was to clone and express the nucleotide sequence of the enzyme which is involved in hyaluronidase enzymatic activity. Materials and Methods: The enzymatic region of hyaluronidase gene was detecte...
متن کاملFunctional and Developmental Analysis of CD4+CD25+ Regulatory T Cells under the Influence of Streptococcal M Protein in Rheumatic Heart Disease
The purpose of this study was to determine the role of streptococcal M protein in naturally-occurring CD4+CD25+ regulatory T cells (nTregs) function and development in rheumatic heart disease in Iraqi patients. Streptococcus pyogenes was isolated for subsequent M protein extraction. Also, peripheral blood nTregs and CD4+ T cells were isolated by using Magnetic Cell Separation System. Tissue cul...
متن کاملReplacement of histidine 340 with alanine inactivates the group A Streptococcus extracellular cysteine protease virulence factor.
Streptococcus pyogenes expresses a highly conserved extracellular cysteine protease that is a virulence factor for invasive disease, including soft tissue infection. Site-directed mutagenesis was used to generate a His340Ala recombinant mutant protein that was made as a stable 40-kDa zymogen by Escherichia coli. Purified His340Ala protein was proteolytically inactive when bovine casein and huma...
متن کاملInactivation of a gene for a fibronectin-binding protein of the oral bacterium Streptococcus mutans partially impairs its adherence to fibronectin.
A sequence of 1647 base pairs in length of Streptococcus mutans DNA that encodes for a 63-kDa protein with significant amino acid similarity with fibronectin-binding proteins of Streptococcus pyogenes and Streptococcus gordonii was cloned. The putative recombinant fibronectin-binding protein of S. mutans was purified using affinity chromatography and the cloned protein was used to prepare polyc...
