چکیده

We describe a new biosensor immunoassay involving optical diffraction to detect clinically Important analytes in human body fluids. A siliconwafer is used as a support for immobilization of antigen or antibody. The protein-coated surface is illuminated through a photo mask to create distinct periodic areas of active and inactive protein. When the surface is incubated with a positive sample, antigen-antibody binding occurs only on the active areas. Upon illumination with a light source such as a laser, the resulting biological diffraction grating diffracts the light. A negative sample does not result in diffraction because no antigen-antibody binding occurs to create the diffraction grating. The presence or absence of a diffraction signal differentiates between positive and negative samples, and the intensity of the signal provides a quantitative measure of the analyte concentration. The technique is demonstrated with a quantitative assay of chonogonadotropin in serum.

A novel optical DNA biosensor for detection of trace concentration of Methylene blue using Gold nano-particles and Guanine rich single strand DNA

The glass surface modification with 3-(mercaptopropyl) trimethoxysilane (MPTS), gold nano-particles (GN) and guanine rich single strand DNA (ss-DNA) was utilized as a novel and efficient platform for sensing trace concentration of methylene blue (MB) by an inexpensive spectrophotometric method. Methylene Blue (MB) can interact with the guanine base of single strand DNA and absorbed onto glass s...

A label-free biosensor-based cell attachment assay for characterization of cell surface molecules

A photonic crystal optical biosensor is incorporated into standard format 96-well microplates for the purpose of detecting the attachment of cells to the biosensor surface without the use of fluorescent labels or any other type of marker. We show that the attachment of cells to the biosensor can be modulated by the immobilization of cognizant ligands to the sensor surface that selectively recog...

Paralytic shellfish poisoning (PSP) toxin binders for optical biosensor technology: problems and possibilities for the future: a review

This review examines the developments in optical biosensor technology, which uses the phenomenon of surface plasmon resonance, for the detection of paralytic shellfish poisoning (PSP) toxins. Optical biosensor technology measures the competitive biomolecular interaction of a specific biological recognition element or binder with a target toxin immobilised onto a sensor chip surface against toxi...
