Nonspecific interferences in the fluorometric determination of plasma cortisol.

چکیده
The popular Mattingly [J. Gun. Pathol. 15, 374 (1962)] fluorometric assay for plasma cortisol is shown to give rise to falsely high values when performed with the recommended optical arrangement. By exciting at 470 nm, however, results are obtained that correlate well (r = 0.972) with those of competitive protein binding. The source of the interference is not identified, but it is related to both the dichloromethane used for extraction and the nature of the individual plasma sample.

Fluorometric determination of plasma 11-hydroxycorticosteroids. II. Studies on the specificity of the method.
We have investigated the specificity of the fluorometric method we proposed [C/in. Chem. 19, 710 (1973)] for plasma or serum li-hydroxycorticosteroid determinations. The principal specific fluorogenic contaminants in the cortisol-containing extract of plasma, as detected by thin-layer chromatography and chemically, were triglycerides and total cholesterol. These contaminants contributed an aver...

A method proposed by Kitabchi and Kitchell [Anal. Biochem. 34, 529 (1970)] for the fluorometric determination of plasma 1 1-hydroxycorticosteroids has been modified. It is simplified by eliminating centrif ugations, by processing all samples consecutively (rather than in small groups), by using disposable test tubes, and by prealkalinizing standards and blanks as well as plasma samples. Specifi...
متن کاملA combined adsorption-gel filtration technique for the determination of the cortisol-binding capacity of transcortin.
A combined adsorption-gel filtration technique has been developed for the quantitation of the cortisol-binding capacity of transcortin: Endogenous steroids are removed from plasma by adsorption on uncoated charcoal. Saturation of the "stripped" binding sites of transcortin is accomplished by equilibrating the sample with a definite amount of labeled cortisol of low specific activity (0.1 muCi/m...
متن کاملDETERMINATION OF SERUM CORTISOL LEVEL BY A DIREC T ENZYME IMMUNOASSAY USING PENICILLINASE AS LABEL
Serum cortisol level was measured by an enzyme immunoassay (EIA) using the enzyme penicillinase as a label without prior extraction and purification. Polyclonal antibodies were raised against cortisol-3-ortho-carboxymethyl-oxime (cortisol-3-0-CMO) conjugated to bovine serum albumin (BSA). This antibody showed a very low cross-reactivity with structurally related steroids (3.7% for corticos...
