چکیده

Intensities of fluorescent images captured by a confocal laser scanning microscope (CLSM) from deep layers of a specimen are often darker than images from the top most layers due to absorption and scattering of both excitation and fluorescent light. These effects cause problems in subsequent analysis of biological objects. Methods [e.g. 1, 2] used for solution of this problem are generally based on the post-processing of data. We have chosen, first, an approach, which lies in software optimization of the microscope hardware during scanning and, second, a postprocessing method based on a simple optimization of image intensities in the image stack.

A New Approach for Scatter Removal and Attenuation Compensation from SPECT/CT Images

Objective(s): In SPECT, the sinogram contains scatter and lack of attenuated counts that degrade the reconstructed image quality and quantity. Many techniques for attenuation and scatter correction have been proposed. An acceptable method of correction is to incorporate effects into an iterative statistical reconstruction. Here, we propose new Maximum Likelihood Expectation Maximiz...

A Quantitative Investigation on the Effect of Edge Enhancement for Improving Visual Acuity at Different Levels of Contrast

Background: The major limitation in human vision is refractive error. Auxiliary equipment and methods for these people are not always available. In addition, limited range of accommodation in adult people when switching from a far point to a near point is not simply possible. In this paper, we are looking for solutions to use the facilities of digital image processing and displaying to improve ...

Compensation of illumination inhomogeneities in multi - resolution image acquisition in confocal microscopy

A multi-resolution application in modern confocal microscopy is based on a set of objectives with different resolution and motorized XY stage. An image captured through a low-resolution objective serves as navigation map where the user points to positions or areas of interest in which a high-resolution image will be captured. This allows e.g. automated acquisition in time lapse experiments, gen...

Variations of the Normal Human Limbal Stem Cells Detected by In Vivo Confocal Microscopy

Background  To report normal variations of the limbal structures using in vivo laser scanning confocal microscopy. Methods: This was a retrospective study of fourteen eyes from 11 healthy individuals. Confocal imaging of cornea and limbus was performed using Heidelberg Retina Tomograph III Rostock Corneal Module. Results: The typical structure of the palisades of Vogt (POV) was detected ...

Adaptive optics confocal microscopy using fluorescent protein guide- stars for brain tissue imaging

Optical aberrations due to the inhomogeneous refractive index of tissue degrade the resolution and brightness of images in deep tissue imaging. We introduce a direct wavefront sensing method using cellular structures labeled with fluorescent proteins in tissues as guide-stars. As a non-invasive and high-speed method, it generalizes the direct wavefront sensing method for adaptive optics microsc...
