چکیده

Abstract: From anesthetized rats in the 90s to conscious wild-type or knockout mice today, intracerebral in vivo microdialysis provided important information about the brain mechanism of action of psychotropic drugs such as antidepressants. The principle of microdialysis technique is based on the balance between the release of neurotransmitters (e.g., serotonin 5-HT, norepinephrine, dopamine) and re-uptake by selective transporters (e.g., SERT for 5-HT). Complementary to electrophysiology, this technique reflects

Extracellular serotonin in the prefrontal cortex is limited through terminal 5-HT1B autoreceptors

Rationale: 5-HT autoreceptors regulate extracellular 5-HT levels and have been suggested to limit the effects of acute treatment with selective serotonin reuptake inhibitors (SSRI). Objectives: The role of terminal 5-HT1B autoreceptors was assessed by comparing the effects of a SSRI on extracellular 5-HT in wildtype and 5-HT1B receptor knockout mice, and by using a 5-HT1B receptor antagonist. S...

In vivo efflux of serotonin in the dorsal raphe nucleus of 5-HT1A receptor knockout mice.

In the dorsal raphe nucleus (DR), extracellular serotonin (5-HT) regulates serotonergic transmission through 5-HT1A autoreceptors. In this work we used in vivo microdialysis to examine the effects of stressful and pharmacological challenges on DR 5-HT efflux in 5-HT1A receptor knockout (5-HT1A-/-) mice and their wild-type counterparts (5-HT1A+/+). Baseline 5-HT concentrations did not differ bet...

The effects of selective serotonin reuptake inhibitors on extracellular 5-HT in the hippo- campus of 5-HT1B receptor knockout mice

The effects of two selective serotonin reuptake inhibitors (SSRIs) on 5-hydroxytryptamine (5-HT) in the hippocampus were studied in wildtype and in 5-HT1B receptor knockout (KO) mice using in vivo microdialysis. Basal 5-HT levels were not different between the two genotypes. The functional absence of 5-HT1B receptors was examined in the knockout mice by local infusion of the 5-HT1B receptor ago...

Effects of chronic paroxetine treatment on dialysate serotonin in 5-HT1B receptor knockout mice.

The role of serotonin (5-HT)1B receptors in the mechanism of action of selective serotonin re-uptake inhibitors (SSRI) was studied by using intracerebral in vivo microdialysis in conscious, freely moving wild-type and 5-HT1B receptor knockout (KO 5-HT1B) mice in order to compare the effects of chronic administration of paroxetine via osmotic minipumps (1 mg per kg per day for 14 days) on extrac...

Antidepressant activity: contribution of brain microdialysis in knock-out mice to the understanding of BDNF/5-HT transporter/5-HT autoreceptor interactions

Why antidepressants vary in terms of efficacy is currently unclear. Despite the leadership of selective serotonin reuptake inhibitors (SSRIs) in the treatment of depression, the precise neurobiological mechanisms involved in their therapeutic action are poorly understood. A better knowledge of molecular interactions between monoaminergic system, pre- and post-synaptic partners, brain neuronal c...
