چکیده

According to current concepts, oxysterols are the physiological mediators of a number of cholesterolinduced metabolic effects (for a review, see Bjorkhem et al1). Most of the evidence for this is however still indirect, and there is a discrepancy between the great number of studies demonstrating potent effects of oxysterols under in vitro conditions and the few studies consistent with a regulatory effect in vivo. Oxysterol binding proteins and receptors are doubtless of regulatory importance in cholesterol turnover, but the physiological ligand(s) to the proteins have not yet been defined with certainty. In this issue of Arteriosclerosis, Thrombosis, and Vascular Biology, the article by Wong et al2 gives strong support for the contention that the oxysterol 24(S),25-epoxycholesterol, an efficient activator of the nuclear receptor LXR in vitro, is an important mediator of statin-induced effects on cholesterol homeostasis in THP-1 macrophages.

24,25-Epoxysterol metabolism in cultured mammalian cells and repression of 3-hydroxy-3-methylglutaryl-CoA reductase.

In view of the potential importance of 24,25-epoxysterols as intracellular regulators of 3-hydroxy-3-methylglutaryl-CoA reductase, the C-24 epimers of 24,25-oxidolanosterol and 24,25-epoxycholesterol were tested for their biological activity and metabolism in cell cultures. All four compounds produced repression of the reductase in cultured mouse fibroblasts (L cells), and both 24(S)- and 24(R)...

Biosynthesis of 24,25-epoxycholesterol from squalene 2,3;22,23-dioxide.

[3H]Squalene 2,3(S);22(S),23-dioxide was prepared by incubation of [3H]mevalonate with S10 rat liver homogenate in the presence of 4,4,10 beta-trimethyl-trans-decal-3 beta-ol, a specific inhibitor of oxidosqualene cyclase (EC 5.4.99.7). Aerobic incubation of [3H]squalene 2,3(S);22(S),23-dioxide with S10 rat liver homogenate affords 24(S),25-epoxycholesterol with an efficiency comparable to that...

CYP3A induction by liver x receptor ligands in primary cultured rat and mouse hepatocytes is mediated by the pregnane X receptor.

The effects of oxysterol and drug ligands of the liver X receptor (LXR) on cytochrome P450 expression were evaluated in primary cultured rodent hepatocytes. Treatment of rat hepatocyte cultures with either 25-hydroxycholesterol or 24(S),25-epoxycholesterol (10(-5) to 5 x 10(-5) M) produced concentration-dependent elevations in CYP3A mRNA and immunoreactive protein levels but did not increase th...

SREBP-2 positively regulates transcription of the cholesterol efflux gene, ABCA1, by generating oxysterol ligands for LXR.

Cholesterol accumulation and removal are regulated by two different transcription factors. SREBP-2 (sterol-regulatory-element-binding protein-2) is best known to up-regulate genes involved in cholesterol biosynthesis and uptake, whereas LXR (liver X receptor) is best known for up-regulating cholesterol efflux genes. An important cholesterol efflux gene that is regulated by LXR is the ATP-bindin...

The Oxysterol 24(S),25-Epoxycholesterol Attenuates Human Smooth Muscle–Derived Foam Cell Formation Via Reduced Low-Density Lipoprotein Uptake and Enhanced Cholesterol Efflux

BACKGROUND Foam cell formation by intimal smooth muscle cells (SMCs) inhibits the elaboration of extracellular matrix, which is detrimental to plaque stabilization. In the present study, we examined the lipoproteins and receptors involved in human SMC foam cell formation and investigated the ability of 24(S),25-epoxycholesterol [24(S),25-EC], an oxysterol agonist of the liver X receptor, to att...
