Prohibitin 2 mRNA downregulation in breast cancer tumor tissues

Authors

Abstract:

Introduction: Breast cancer is the most important cause of cancer mortality among women, therefore the study of its causative or aggravating factors seems necessary. In this study, the mRNA levels of the PHB2 gene were evaluated in tumor and adjacent non-tumor tissues of 50 women diagnosed with invasive ductal carcinoma of the breast. Methods: RNX-Plus solution was used to isolate total RNA from tumor and adjacent non-tumor tissues of breast cancer patients. Thereafter, total RNA was converted to cDNA using PrimeScriptTM RT reagent kit. The mRNA levels of PHB2 were quantified by qRT-PCR and data were analyzed with a paired-samples t test. Furthermore, ROC curve analysis was performed to evaluate the diagnostic capacity of PHB2 expression in breast cancer tumor tissues. Results: A significantly lower level of PHB2 mRNA was observed in tumor tissues of breast cancer patients compared with adjacent non-tumor tissues (P<0.0001). PHB2 mRNA levels showed an approximately 2.5-fold reduction in tumor tissues compared with normal tissues (P = 0.001). Roc cure analysis showed that PHB2 mRNA level can discriminate tumors from non-tumor tissues with 91.3% specificity and 64.3% sensitivity (AUC=0.712, P<0.001). Conclusion: Downregulation of PHB2 in breast cancer patients shows that its mRNA levels can possibly discriminate tumors from non-tumor tissues.

Downregulation of lung mitochondrial prohibitin in COPD.

Prohibitins (PHB1 and PHB2) are versatile proteins located at the inner mitochondrial membrane, maintaining normal mitochondrial function and morphology. They interact with the NADH dehydrogenase protein complex, which is essential for oxidoreductase activity within cells. However, their expression in lung epithelium, especially in smokers and patients with inflammatory lung diseases associated...

Detection of Toxoplasma gondii DNA in Malignant Breast Tissues in Breast Cancer Patients

Breast cancer is the most prevalent malignancy in women throughout the world. Similar to other cancers, a strong relationship between breast cancer and environmental factors such as infectious agents has been reported. Toxoplasma gondii is a protozoan parasite which may play a role in cancer induction. The present study aimed to investigate a possible association between a history of T. gondii ...

Tumor suppression by the prohibitin gene 3'untranslated region RNA in human breast cancer.

Prohibitin is a candidate tumor suppressor gene located on human chromosome 17q21, a region of frequent loss of heterozygosity in breast cancers. We showed previously that microinjection of RNA encoded by the prohibitin gene 3'untranslated region (3'UTR) blocks the G(1)-S transition causing cell cycle arrest in several human cancer cell lines, including MCF7. Two allelic forms (C versus T) of t...

Nerve Fibers in Breast Cancer Tissues Indicate Aggressive Tumor Progression

Emerging evidence has indicated nerve fibers as a marker in the progression of various types of cancers, such as pancreatic cancer and prostate cancer. However, whether nerve fibers are associated with breast cancer progression remains unclear. In this study, we evaluated the presence of nerve fibers in 352 breast cancer specimens and 83 benign breast tissue specimens including 43 cases of cyst...

CTGF and MLH1 Gene Expression Levels in Colorectal Cancer Tumor Tissues and Adjacent Normal Tissues in Patients in Golestan Province

Background and purpose: Colorectal cancer is the third most common type of cancer in terms of incidence and the second most common cause of cancer-related death worldwide. The aim of this study was to investigate the expression of CTGF and MLH1 gene in colorectal cancer tumor tissues and adjacent normal tissues in patients in Golestan province. Materials and methods: In this experimental study...

Prohibitin 3' untranslated region polymorphism and breast cancer risk.

Introduction Prohibitin is an antiproliferative protein (1) and may function as a tumor suppressor through interaction with the retinoblastoma tumor suppressor protein and its family members (2). In addition, somatic mutations have been identified in human cancers, including breast cancer (3, 4). Interestingly, the prohibitin 3 UTR exhibits the characteristics of a trans-acting regulatory RNA t...
