the influence of corpus luteum on follicular fluid composition from different size follicles and their relationship to serum concentrations in sanjabi ewes

abstract

the aim of the present study was to determine the in?uence of presence or absence of corpus luteum (cl) on hormonal and metabolites composition of follicular ?uid (ff) harvested from different sized follicles and its relationship with blood serum concentrations in sanjabi ewes. ovaries and blood samples were collected from 60 clinically healthy adult ewes (sanjabi breed) 1–3 years of age in dioestrous period of the estrus cycle. the ovaries collected per ewe were classi?ed as with corpus luteum (cl+) and without corpus luteum (cl-). follicular ?uid was aspirated from small (1 to 4mm) and large (?4.1 mm) follicles in cl+ and cl- ovaries. follicular ?uid (ff) was aspirated from different sized follicles in cl+ and cl- ovaries. serum and ff samples were analyzed for hormones (estradiol-17?, progesterone and testosterone) and biochemical metabolites (glucose, cholesterol, triglyceride, total protein, albumin and globulin). results showed that the ff concentration of estradiol-17?, progesterone, testosterone and albumin in different sized follicles categories (small and large follicles in cl+ and cl- ovaries) were signi?cantly higher than those in the serum (p<0.05). while the concentrations of glucose, cholesterol, triglyceride and globulin in the serum was significantly higher when compared with the ?uid from different sized follicles categories (small and large follicles in cl+ and cl- ovaries). in the cl+ and cl- ovaries, estradiol-17?, progesterone, glucose and cholesterol concentrations increased significantly as follicle size increased (p<0.05). the ff concentration of testosterone, estradiol-17?, glucose, triglyceride and cholesterol in same follicle size categories in cl+ and cl- ovaries were also signi?cant (p<0.05). note that, concentration of these hormones and metabolites in the cl+ ovaries were lower than that of the cl- ovaries. the ff concentration of progesterone in large follicle size category in cl+ ovaries was signi?cantly higher than that of the cl- ovaries (p<0.05). there was a positive signi?cant correlation between concentration of progesterone in serum and ff from different sized follicles in cl+ ovaries (p<0.01). also, there was a signi?cant positive correlation between concentration of glucose and total protein in serum and ff from different sized follicles in cl- ovaries (p<0.01).in conclusion, the levels of hormones and metabolite composition in the ff interestingly were related to presence or absence of a cl and also to the follicular size.
