چکیده مقاله:

Background and Objective: Chronic arsenic toxicity is a widespread problem; the role of brain oxidative stress has been suggested in the genesis of epilepsy and in the post-seizure neuronal death. However, studies investigating the effects of arsenic on seizure and related mechanisms are limited. The purpose of this study was to examine the effect of prolonged exposure to sodium arsenite on oxidative damage in pentylenetetrazole (PTZ)-induced seizures in mice. Materials and Methods: In this study, male NMRI mice received sodium arsenite (0, 25, 50, and 100 ppm) in the drinking water for a period of 30 days. After exposure, all animals were injected PTZ (PTZ; 85 mg/kg, i.p.) to induce seizure, and the seizure parameters were evaluated for 30 minutes. Then, the levels of malondialdehyde (MDA) and reduced glutathione (GSH) were measured in the brain.   Results: The results of this study showed that sodium arsenite decreases the latency to the seizure onset and time of death (p

Positive effects of Vitamin C in arsenic trioxide and sodium fluoride induced genotoxicity and oxidative stress in mice in vivo

The aim of the present study was to evaluate Vitamin C (VC) as a potent natural antioxidant to mitigate the genotoxic effects of Arsenic trioxide and sodium fluoride in Swiss albino mice in vivo. The study was divided into eight groups consisting of control treated with normal saline (Group I), Group II, III, IV, V, VI, VII and the VC group with only Vitamin C (500 mg/kgbw). Arsenic trioxide (4...

Effect of Ocimum basilicum hydro-alcoholic extract on oxidative damage of brain tissue following seizures induced by pentylenetetrazole in mice

Introduction: A relationship between epileptic seizures and brain tissue oxidative damage has been suggested. Ocimum basilicum (O. basilicum) has been shown to have beneficial effects including hypnotic and protective against tissue oxidative damage. The present study was designed to evaluate the effects of O. basilicum hydro-alcoholic extract on oxidative damage of brain tissue following s...

Evaluation of Anticonvulsant Effects of Nimodipine and Ascorbic Acid on Pentylenetetrazole- Induced Seizures in Mice

Introduction: The mechanisms underlying the vulnerability of the brain to seizures remains unknown. Calcium ions influx and oxidative stress have been implicated in a variety of acute and chronic epileptogenic conditions. The present study was aimed at investigating the effects of nimodipine and ascorbic acid (vitamin C) alone and in combination on pentylenetetrazole (PTZ) - induced seizures ...

Pharmacological Studies on Khamira Marwarid: Effect on Pentylenetetrazole-Induced Seizures, Cognition and Biochemical Markers in Mice

Cognitive impairment in epileptics may be a consequence of the epileptogenic process as well as antiepileptic medication. Thus, there is need for drugs, which can suppress epileptogenesis as well as prevent cognitive impairment. In the present study, the effect of Khamira Marwarid (KAH 1 ), a formulation based on Indian system of Unani medicine, was evaluated on the course of pentylenetetrazole...

Effects of neuropeptide S on seizures and oxidative damage induced by pentylenetetrazole in mice

Neuropeptide S (NPS) and its receptor were recently discovered in the central nervous system. In rodents, NPS promotes hyperlocomotion, wakefulness, anxiolysis, anorexia, and analgesia and enhances memory when injected intracerebroventricularly (i.c.v.). Herein, NPS at different doses (0.01, 0.1 and 1nmol) was i.c.v. administered in mice challenged with pentylenetetrazole (PTZ; 60mg/kg) repeate...
