Changes of miR-199a and miR-196b expression levels in blood donors suffering from asymptomatic hepatitis C

چکیده مقاله:

Abstract Background and Objectives Hepatitis C virus is the main cause of hepatitis C infection that usually has no symptoms at early stages. Studies have shown changes in the expression level of microRNAs in various diseases such as hepatitis. MiR-199a and miR-196b have an inhibitory effect on hepatitis C virus. In a recent study, we examined the expression levels of miR-199a and miR-196b in serum samples of asymptomatic hepatitis C infected blood donors in comparison to serum samples of healthy blood donors; based on the results, the importance of microRNAs in different stages of hepatitis C infection was evaluated.   Materials and Methods In this case control study, first total RNA extraction from 106 serum samples of healthy blood donors (control group) and 106 serum samples of asymptomatic hepatitis C infected blood donors (case group) was carried out. After cDNA synthesis, the expression level of miR-199a and miR-196b were evaluated by SYBR Green real-time PCR, and the results were analyzed by REST 2009 software.   Results Expression level of miR-196b in the case group compared to the control group was lower (p 1clip_image001.png" >  0.001). No statistically significant difference was observed in the expression level of miR-199a between the two groups.   Conclusions  With regard to lower expression level of miR-196b, and given no significant difference of miR-199a expression level in the case group in comparison to the control group, future studies on the role of miR-196b in this disease are required.

Hepatitis C virus infection and genotypes in blood donors

Abstract: Genotyping of the hepatitis C virus (HCV) is important for designing therapeutic strategies and regional specific diagnostic assays. The aim of this study was to identify the HCV genotypes in HCV infected blood donors. This is the first report on HCV genotypes in blood donors in Iran. In this cross-sectional study, 103 blood donors with hepatitis C were investigated for HCV genotype...

Differential expression of miR-1297, miR-3191-5p, miR-4435, and miR-4465 in malignant and benign breast tumors

Objective(s): MicroRNAs (miRs) are a class of small non-coding RNAs which are associated with tumor growth and progression. In the present study, we assessed the expression of selected miRs in malignant, benign, and adjacent normal breast tissues. Materials and Methods: The expression of miR-1297, miR-3191-5P, miR-4435, and miR-4465 were...

Evaluation of miR-141, miR-200c, miR-30b Expression and Clinicopathological Features of Bladder Cancer

Bladder cancer (BC) ranks the second most common genitourinary tract malignant tumor with high mortality and 70% recurrence rate worldwide. MiRNAs expression has noticeable role in bladder tumorigenesis. The purpose of this study was to assess miR-200c, miR-30b and miR-141 in tissue samples of patients with BC and healthy adjacent tissue samples and their association with muscle invasion, grade...

Expression Analysis of mir-21 and mir-221 in Cancerous Tissues from Iranian Patients with Gastric Cancer

Background: Early detection is a key to survival for gastric cancer. Molecular markers such as miRNA (microRNA) can have great importance in the early diagnosis of gastric cancer. Expression of miR-21 and miR-221 are deregulated in many types of human cancers. This study aimed to investigate the differences in miRNA expression patterns within the Iranian population. Methods: Total RNA was extra...
