چکیده مقاله:

Objectives- To assess the ectopic bone formation in canine masseter muscle following the implantation of the natural bovine bone minerals (NBM) loaded with canine mesenchymal stem cells (MSCs).Design- Experimental study.Animals- four mongrel dogs.Procedures- Tripotent MSCs isolated from the canine bone marrow were loaded onto the NBM sponges and allowed to adhere. The cell-loaded scaffolds were then implanted in parallel with cell-free control scaffolds in masseter muscles of four mongrel dogs. Eight weeks after, the animals were sacrificed and the ectopic bone formation in implantation site was studied using the sections prepared from the parts of the muscle containing the implants. Furthermore, the amount of bone formation in two studied groups was quantified using Image-Pro Plust software.Results- The implants from the both groups were appeared to be encapsulated by fibrous tissue in implantation site which included some trabecular bone containing osteocyte and osteoblast. There were no indications of inflammation and foreign body reaction, nor were there any indications of cartilage tissue formation. In contrast to control, in MSCs group, lamellar bone was observed in some area. More importantly, in cell loaded scaffolds more amount of bone was formed compared to that of control cell free scaffolds (P<0.05).Conclusion and Clinical Relevance- Taken together it seems that in vivo bone forming capacity of the NBM sponges would be improved by loading it with MSCs.

Capillary Network Formation by Endothelial Cells Differentiated from Human Bone Marrow Mesenchymal Stem Cells

Human bone marrow derived mesenchymal stem cells (HBMSCs) have the potential to differentiate into cells such as adipocyte, osteocyte, hepatocyte and endothelial cells. In this study, the differentiation of hBMSCs into endothelial like-cells was induced in presence of vascular endothelial growth factor (VEGF) and insulin-like growth factor (IGF-1). The differentiated endothelial cells were exam...

Isolation, characterization and transduction of canine bone marrow-derived mesenchymal stem cells (cBM-MSCs)

BACKGROUND: Stem cell therapy in small animal medicineis still in its infancy and few in vitro and in vivo research projectsregarding animal Mesenchymal Stem Cells (MSCs) have beencarried out. On the other hand, Cell tracking is the first step of thecell-based therapies and is essential to recognize cell fate posttransplantation. OBJECTIVES: The aim of this study was toisolate, characterize, an...

Harvesting of bone marrow mesenchymal stem cells from live rats and the in vitro differentiation of bone marrow mesenchymal stem cells into neuron-like cells

In the bone marrow, there are certain populations of stem cell sources with the capacity to differentiate into several different types of cells. Ideally, cell transplants would be readily obtainable, easy to expand and bank, and capable of surviving for sufficient periods of time. Bone marrow mesenchymal stem cells (BM-MSCs) possess all of these characteristics. One of the most important benefi...

Ectopic Bone Formation by Mesenchymal Stem Cells Derived from Human Term Placenta and the Decidua

Mesenchymal stem cells (MSCs) are one of the most attractive cell types for cell-based bone tissue repair applications. Fetal-derived MSCs and maternal-derived MSCs have been isolated from chorionic villi of human term placenta and the decidua basalis attached to the placenta following delivery, respectively. Chorionic-derived MSCs (CMSCs) and decidua-derived MSCs (DMSCs) generated in this stud...
