چکیده مقاله:

This study was performed in order to define the cellular immune response of 12 recovered cutaneous leishmaniasis subjects to different soluble antigens of the amastigote form of Leishmania major. A soluble leishmanial antigen preparation (SLA) derived from highly-purified amastigotes from infected nude mice was fractionated by Mono Q column using Fast Protein Liquid Chromatography (FPLC) system. Three different fractions were obtained. The lymphoproliferative response, interferon•gamma (IFN.'Y) and interleukin•4 (JL•4) to amastigote SLA and its three subfractions were measured. The highest proliferative response and specific IFN.'Y production without synthesis of fL•4, was induced by the fITst fraction of amastigote SLA. These results showed that the individuals who had recovered from cutaneous leishmaniasis had expanded memory T- cell clones recognizing different antigens in the first fraction of amastigote SLA. The main and most important point of this study was the identification of one fraction of an amastigote antigen which preferentially reacted with cells from recovered human cutaneous leishmaniasis cases.

Leishmania pifanoi amastigote antigens protect mice against cutaneous leishmaniasis.

In the search for a leishmaniasis vaccine, extensive studies have been carried out with promastigote (insect stage) molecules. Information in this regard on amastigote (mammalian host stage) molecules is limited. To investigate host immune responses to Leishmania amastigote antigens, we purified three stage-specific antigens (A2, P4, and P8) from in vitro-cultivated amastigotes of Leishmania pi...

Cationic Immune Stimulating Complexes Containing Soluble Leishmania Antigens: Preparation, Characterization and in Vivo Immune Response Evaluation

Background: Cationic immune stimulating complexes (PLUSCOMs) are particulate antigen delivery systems. PLUSCOMs consist of cationic immunostimulatory complexes (ISCOMs) derivatives and are able to elicit in vivo T cell responses against an antigen. Objective: To evaluate the effects of PLUSCOMs containing Leishmaniamajor antigens (SLA) on the type of immune response generated in the murine mod...

Sphingomyelin Liposomes Containing Soluble Leishmania major antigens Induced Strong Th2 Immune Response in BALB/c Mice

Objective(s): Soluble Leishmania antigens (SLA) provide suitable protection against leishmaniasis in murine model when delivered by an appropriate delivery system. Liposomes have been shown to be suitable vaccine delivery systems against leishmaniasis, however, the phospholipase-A (PLA) activity of SLA is a drawback to prepare a stable liposomal SLA. One strategy to overcome this problem migh...

Immune responses to soluble antigens of treponemes.

Sonicates from five cultivable treponemes were used as antigens in delayed hypersensitivity tests and macrophage inhibition assays. Immunodiffusion analysis showed that the sonicate comprised two major antigenic components which were not distinguishable in the skin tests. The sonicate antigens elicited significant cell-mediated immunity in guinea-pigs. Treponema refringens biotype Nichols prove...
