چکیده مقاله:

Because of expanding resistance to efficient and affordable antimalarial drugs likechloroquine, the search is continuing for more effective drugs against this disease. In-vitroantiplasmodial activity and cytotoxicity of α-(acyloxy)-α-(quinolin-4-yl) acetamides onPlasmodium falciparum and structure-activity relationships of this new class of Passeriniadducts is described. The in-vitro antiplasmodial activity of compounds was tested againstchloroquine sensitive 3D7 strain. Toxicity of active compounds was investigated on HepG2cell line. Compounds 1, 20 and 22 showed significant antiplasmodial activity with IC50 valueof 1.511, 1.373 and 1.325 μM, respectively. The active compounds did not show noticeabletoxicity when tested against HepG2 cell line. The present results bring essential elementswhich will be used for the synthesis of more active derivatives of α-(acyloxy)-α-(quinolin-4-yl)acetamides.

Evaluation of α,β-unsaturated ketones as antileishmanial agents.

In this study, we assessed the antileishmanial activity of 126 α,β-unsaturated ketones. The compounds NC901, NC884, and NC2459 showed high leishmanicidal activity for both the extracellular (50% effective concentration [EC50], 456 nM, 1,122 nM, and 20 nM, respectively) and intracellular (EC50, 1,870 nM, 937 nM, and 625 nM, respectively) forms of Leishmania major propagated in macrophages, with ...

SAR and identification of 2-(quinolin-4-yloxy)acetamides as Mycobacterium tuberculosis cytochrome bc1 inhibitors.

A previous phenotypic screen by GSK identified 2-(quinolin-4-yloxy)acetamides as potent growth inhibitors of Mycobacterium tuberculosis (Mtb). We report the results of a preliminary structure-activity relationship (SAR) study of the compound class which has yielded more potent inhibitors. An Mtb cytochrome bd oxidase deletion mutant (cydKO) was found to be hypersensitive to most members of the ...

Synthesis and Evaluation of α-Thymidine Analogues as Novel Antimalarials

Plasmodium falciparum thymidylate kinase (PfTMPK) is a key enzyme in pyrimidine nucleotide biosynthesis. 3-Trifluoromethyl-4-chloro-phenyl-urea-α-thymidine has been reported as an inhibitor of Mycobacterium tuberculosis TMPK (MtTMPK). Starting from this point, we designed, synthesized and evaluated a number of thymidine analogues as antimalarials. Both 5'-urea-α- and β-thymidine derivatives wer...

Do α-acyloxy and α-alkoxycarbonyloxy radicals fragment to form acyl and alkoxycarbonyl radicals?

The generation of alpha-acyloxy and alpha-alkoxycarbonyloxy radicals under reductive conditions in fragmentable probe experiments does not provide unequivocal evidence for the fragmentation of such radicals to give ketones and acyl or alkoxycarbonyl radicals. Instead, standard reduction predominates, even at low tin hydride concentrations. Some ketone product is formed in the alpha-acyloxy subs...
