چکیده مقاله:

Several studies have demonstrated that nitric oxide (NO) is involved in hyperalgesia induced by NMDA activity in Morphine tolerance and NO synthase inhibitors can attenuate hyperalgesia in morphine tolerance. In addition, one of the possible mechanisms in morphine tolerance is reduction of GABA inhibitory effect. For this reason, we used GABA agonists (muscimol and baclofen) to investigate the effect of GABA agonists on analgesic effect of morphine and NO level of serum. In this study, experimental rats were divided into 4 groups as follows: 1. Control group, 2. Morphine tolerance group, 3. Morphine tolerance + muscimol, and 4-morphine tolerance + baclofen. To induce morphine tolerance in rats, the animals received 10 mg/kg of morphine sulfate intraperitoneal (ip) once a day for 8 days. In the treatment group, GABA agonist was injected on the 1st, 3rd, 5th, and 8th day before morphine injection. Finally, to evaluate GABA agonists treatment on morphine analgesia and hyperalgesia on the 8th day of the study, thermal hyperalgesia was used to evaluate tolerance and hyperalgesia. Then, NO level of serum was investigated as an important factor in OIH. The results indicated that GABA agonists reduced thermal hyperalgesia in the morphine tolerance group (p

Effect of sub-effective dose of GABA agonists on attenuation of morphine tolerance in rats: Behavioral and electrophysiological studies

GABAergic drugs can change analgesic effect of morphine. Wide dynamic range (WDR) neurons play an important role in pain transmission and may change behaviors in morphine tolerance. In this study WDR neuron behaviors in morphine tolerant rats and rats treated with GABA agonists, were recorded to elucidate the effect of morphine and GABA agonists on WDR behavioral changes. Rats were divided to 4...

Cellular Mechanisms Underlying Morphine Analgesic Tolerance and Hyperalgesia

Opioid dose escalation or analgesic tolerance is observed during longer treatments in a significant number of patients with chronic pain owing to cancer or nonmalignant tissue injury. Higher doses of morphine are more likely to result in subsensitivity to the drug and worsened quality of life (QOL) by exerting other side effects. Many investigators have been studying the molecular and cellular ...

Mesenchymal Stem Cells Reversed Morphine Tolerance and Opioid-induced Hyperalgesia

More than 240 million opioid prescriptions are dispensed annually to treat pain in the US. The use of opioids is commonly associated with opioid tolerance (OT) and opioid-induced hyperalgesia (OIH), which limit efficacy and compromise safety. The dearth of effective way to prevent or treat OT and OIH is a major medical challenge. We hypothesized that mesenchymal stem cells (MSCs) attenuate OT a...

Complement factor C5a and C5a receptor contribute to morphine tolerance and withdrawal-induced hyperalgesia in rats

Morphine is a potent opioid analgesic. However, the repeated use of morphine causes tolerance and hyperalgesia. Neuroinflammation has been reported to be involved in morphine tolerance and withdrawal-induced hyperalgesia. The complement system is a crucial effector mechanism of immune responses. The present study investigated the roles of complement factor C5a and C5a receptor (C5aR) in the dev...
