چکیده مقاله:

Background: Mycobacterium bovis is the main cause of tuberculosis in cattle. At the global scale and also in Iran, the most frequent currently-in-use method in the detection of infected cattle is tuberculination. Objective: The present study was aimed to improve our genomic knowledge of Mycobacterium bovis population structure in cattle farms of Shiraz. Materials and Methods: Fifty pathological samples from tuberculin-positive cattle collected from two slaughterhouses at Shiraz were subjected to bacterial culture on glycerinated and pyruvated Lowenstein-Jensen media. Genomic material from culture-positive slopes was extracted and used in PCR-16S rRNA, PCR-IS6110, and PCR-RD typing experiments. All the M. bovis isolates were further genotyped using pvu II-digested PGRS-RFLP strategy. Results: In bacterial culture, a total of 13 (26%) samples proved to carry live mycobacteria where in PCRs their identity was all shown to be M. bovis. Genotype profiling by RFLP-PGRS displayed two patterns with 10 isolates shared a single profile identical to that of M. bovis BCG (1173 P2) strain and three holding a different genotype. Conclusion: While higher frequency of a BCG-like M. bovis in cattle farms of Shiraz is not surprising as this is a typical characteristic of Iranian M. bovis population, we assumer detection of a highly similar strain found in our work might indicate local evolution of new M. bovis strains in the region or infiltration of such strain(s) through cattle farming activities.

Molecular characterization of Mycobacterium bovis isolates from Ethiopian cattle

BACKGROUND Bovine Tuberculosis (BTB) is a widespread and endemic disease of cattle in Ethiopia. Information relating to genotypic characteristics of Mycobacterium bovis strains affecting the cattle population in Ethiopia is limited. We carried out molecular characterization of M. bovis strains isolated from BTB infected cattle using the spoligotyping technique. The relationship between distribu...

Molecular and genomic features of Mycobacterium bovis strain 1595 isolated from Korean cattle

The aim of this study was to investigate the molecular characteristics and to conduct a comparative genomic analysis of Mycobacterium (M.) bovis strain 1595 isolated from a native Korean cow. Molecular typing showed that M. bovis 1595 has spoligotype SB0140 with mycobacterial interspersed repetitive units-variable number of tandem repeats typing of 4-2-5-3-2-7-5-5-4-3-4-3-4-3, representing the ...

Molecular identification of Mycobacterium bovis from cattle and human host in Mali: expanded genetic diversity

BACKGROUND Bovine tuberculosis (BTB) is a contagious, debilitating human and animal disease caused by Mycobacterium bovis, a member of the Mycobacterium tuberculosis complex. The study objective were to estimate the frequency of BTB, examine genetic diversity of the M. bovis population in cattle from five regions in Mali and to determine whether M. bovis is involved in active tuberculosis (TB) ...

Isolation of Mycobacterium bovis & M. tuberculosis from cattle of some farms in north India--possible relevance in human health.

BACKGROUND & OBJECTIVE Infection due to Mycobacterium bovis typically occurs in cattle and animals transmit infection to each other. The choice of appropriate clinical specimen is very important for isolation of M. bovis and M. tuberculosis from cattle. The present study reports the isolation of M. tuberculosis and M. bovis from different types of specimens from cattle suspected to be suffering...
