چکیده مقاله:

Objective Articular cartilage as an avascular skeletal tissue possesses limited capacity to heal. On the other hand, it is believed that the regeneration capacity of each tissue is largely related to its stem cell contents. Little is known about the presence of mesenchymal stem cells in articular cartilage tissue. This subject is investigated in the present study. Materials and Methods Articular cartilage was obtained from rats of Wistar strain and subjected to enzymatic digestion using 1:1 ratio of 0.2% collagenase I and 0.1% pronase. The released cells were then collected and cultivated in        25 cm2 culture flask, which was resulted in the appearance of two morphologically distinct cell populations. Fibroblastic population were purified and expanded through several subcultures, using their loose adherence on culture surfaces and examined in terms of differentiation potential. Results The primary culture mainly contained polygonal cells with few fibroblastic cells among them. Upon the first subculture, the fibroblastic cells appeared as aggregates of few cells among the polygonal cartilage cells. It was found that the fibroblastic cell population could easily detach from the culture surface following to a brief exposure to tripsin solution. Using this property, these cells purified and expanded by several subcultures. According to the results, the isolated cells were able to easily differentiate into chondrocytic, osteocytic and adipocytic cell lineages. Conclusion It seems that articular cartilage contains a distinct fibroblastic cell population with mesenchymal stem cell nature. These cells can be purified and expanded by cell culture methods, thanks to their loose adherence on the culture dish surfaces.

Articular cartilage stem cell signalling

The view of articular cartilage as a non-regeneration organ has been challenged in recent years. The articular cartilage consists of distinct zones with different cellular and molecular phenotypes, and the superficial zone has been hypothesized to harbour stem cells. Furthermore, the articular cartilage demonstrates a distinct pattern regarding stem cell markers (that is, Notch-1, Stro-1, and v...

Conditioned medium derived from mesenchymal Stem cells regenerates’ defected articular cartilage

Background & Aims: One of cell- based technical issues associated with cartilage repair assay is delivering cells to the site of the parts where damage is created. Mesenchymal stem cells (MSCs) with their chondrogenic potential are ideal candidates for cartilage regeneration. High expression of cartilage hypertrophy markers by MSCs would result in apoptosis and ossification. This investigation ...

Study of Differentiation Potential of the Dedifferentiated Chondrocytes From Rat Articular Cartilage into Skeletal Cell Lineages

Purpose: Dedifferentiation of the chondrocyte from rat articular cartilage with multiple subcultures and study of the redifferentiation potential of the cells into bone, cartilage and fat cell lineages. Materials and Methods: In this experimental study, chondrocytes from rat articular cartilage were isolated and expanded through several successive subcultures during which the expression levels ...

Mesenchymal stem cells can survive on the extracellular matrix-derived decellularized bovine articular cartilage scaffold

Objective (s): The scarcity of articular cartilage defect to repair due to absence of blood vessels and tissue engineering is one of the promising approaches for cartilage regeneration. The objective of this study was to prepare an extracellular matrix derived decellularized bovine articular cartilage scaffold and investigate its interactions with seeded rat bone marrow mesenchymal stem cells (...

Development and Application of Mesenchymal Stem Cell-derived Exosomes in Cartilage Tissue Repair

Background and Aim: Cartilage defects treatment is one of the most common clinical challenges in orthopedics. The current management techniques help to control symptom and joint function. The cell-free approach to cartilage regeneration through paracrine action has been considered to accelerate and facilitate the healing process and the importance of its urgency in the recovery of military pe...
