چکیده مقاله:

Background: Low uptake of exogenous DNA by sperm and reduced number of fertilized oocyte by transfected sperm are the major obstacles for progression of sperm mediated gene transfer. Therefore, the modification of sperm mediated gene transfer procedure needs to be required. The purpose of this study was to evaluate the efficiency of FuGene 6 compare to lipofection in transfection medium for internalizing of the foreign gene in the rooster sperm cell. Materials and Methods: Semen were washed in Beltsville extender for removal of the seminal plasma by repetitive washing. Then sperm (1 ×109 sperm/ml) was incubated with exogenous DNA (Enhanced Green Fluorescent Protein, EGFP) in Beltsville extender containing: 1. no additive, 2. lipofectamine 1000 (1:1), and 3. FuGene 6 (1:1). Transfection was measured using PCR reaction in the sperm cell and new born chickens after artificial insemination. Motility, viability, membrane integrity, acrosome integrity and their fertilizing ability using artificial insemination were considered as index of sperm quality post transfection. Results: Although there was not significant different in rate of transfection in sperm cell between Lipofectamin (11%) and FuGene 6 (10%), they were significantly higher compare to Control group (2%). The higher significant percentage of motility, viability and fertility rate were observed in FuGene 6 (29.1 ± 1.5, 33.5 ± 2.4, 26.8 ± 1.8, respectively) compare to control group (10.5 ± 1.5, 12.8 ± 2.4, 14.9 ± 1.8) and Lipofectamine (13.5 ± 1.5, 16.1 ± 2.4, 17.3 ± 1.8). Moreover, the higher significant percentage of transfection in chickens were observed in FugGen 6 (33.5%) rather than control (14%) and lipofectamine (16%) groups. For membrane integrity and acrosome integrity, no significant different was observed for all groups. Conclusion: Our data revealed that adding FuGene 6 would resulte in higher rate of sperm quality after transfection which led to higher rate of fertility and production of transgenic chicken.

Sperm Mediated Gene Transfer (SMGT) in Chicken

A transgenic animal as originally defined by Gordon and Ruddle (1981), is an organism carrying recombinant molecules that were intentionally introduced by human intervention. Classically this entailed the stable integration of the transgene within the host animal’s genome and its transmission to progeny through normal breeding programmes. Transgenic chicken holds great promise in basic biologic...

Sperm-mediated gene transfer.

Since 1989, a new method for the production of transgenic animals has been available, namely sperm-mediated gene transfer (SMGT), based on the intrinsic ability of sperm cells to bind and internalise exogenous DNA molecules and to transfer them into the oocyte at fertilisation. We first described the SMGT procedure in a small animal model, with high efficiency reported in the mouse. In addition...

P-85: Epididymal Sperm Parameters following Sperm Mediated Gene Transfer of EDFP-pIRES Vector in Rat

Background: Sperm mediated gene transfer was approved for producing transgenic animals. There are many studies which reported different success rate by this procedure. Failure in this project mostly is returned to transfection and stability of transferred genes. The other problem that may contribute in project failure may return to the testicular damages which occur at the time of transfection,...

Production of transgenic mice expressing tumor virus A under ovarian‑specific promoter 1 control using testis‑mediated gene transfer.

The aim of the present study was to produce transgenic mice expressing tumor virus A (TVA) in the ovary under ovarian specific promoter 1 (OSP1) control. A transgenic mouse model was established in which TVA, an avian retroviral receptor gene driven by OSP1, was selectively expressed in the ovary. A recombinant plasmid containing TVA cDNA and an OSP1 promoter was constructed. The DNA fragment w...

Efficient production by sperm-mediated gene transfer of human decay accelerating factor (hDAF) transgenic pigs for xenotransplantation.

A large number of hDAF transgenic pigs to be used for xenotransplantation research were generated by using sperm-mediated gene transfer (SMGT). The efficiency of transgenesis obtained with SMGT was much greater than with any other method. In the experiments reported, up to 80% of pigs had the transgene integrated into the genome. Most of the pigs carrying the hDAF gene transcribed it in a stabl...

Production of Transgenic Goats by Sperm-mediated Exogenous DNA Transfer Method*

In this study, the production of transgenic goats using sperm to integrate exogenous DNA and artificial insemination (AI) was carried out and the technical protocols for sperm-mediated gene transfer (SMGT) in the goat were optimized. The standard sperm parameters and the ability to bind foreign genes were assessed to select suitable sperm donor bucks. A total of 134 oestrous does were divided i...
