Prevalence of anti-HBc in HBsAg negative blood donors using two enzyme immunoassays kits

چکیده مقاله:

Abstract Background and Objectives Use of kits with high sensitivity and specificity to determine the prevalence of antibodies against hepatitis B virus (anti-HBc) core antigen is important. The aim of this study was to determine the prevalence of hepatitis B virus antibody in HBsAg negative blood donors using two anti-HBc enzyme immunoassay kits (DIA.PRO and SIEMENS).   Materials and Methods In this Cross-sectional study, a number of 4313 samples of HBsAg negative blood donors were collected. All samples were tested for the detection of anti-HBc (Total, IgM & IgG) by ELISA using Diapro and Siemens kits. Anti-HBc assay was performed by both kits in duplicate. Statistical analysis of data was performed using SPSS Ver. 22.0; chi-square tests were also performed.   Results The study population consisted of 227 (5.3%) women and 4086 (94.7%) men. 498 (11.5%) and 384 (8.9%) samples reacted with Diapro and Siemens kits for Anti-HBc, respectively. All positive samples of both kits in duplicate test showed the same results as the previously reported results. 114 cases of reactive kits of DIA.PRO kit were negative in re-testing with SIEMENS kit. Significant differences were observed between the reactive results of the two kits (p = 0.00005).  The kappa coefficient for the reactive results of the two kits was 85%.   Conclusions  Considering the significant difference between the results of the two kits, it is recommended to perform anti-HBc testing with anti-HBs and /or anti-HBe to determine the prevalence of anti-HBc in blood donors with past HBV infection.

Detection of HBV DNA in HBsAg Negative Normal Blood Donors

Background: The risk of infection by transfusion-transmitted viruses has been reduced remarkably. However, a zero-risk blood supply is still desirable. The screening for antibody to HBc (anti-HBc) has been shown as an alternative test for the detection of HBV infection. Objective: The main aim of this study was to evaluate HBV infection markers and the potential value of anti-HBc testing of blo...

Prevalence of Occult Hepatitis B Infection among HBsAg Negative Blood Donors in Golestan Province

Background and Aims: Occult hepatitis B virus infection (OBI) is known as an important source of hepatitis B virus (HBV) infection. It is categorized as Hepatitis B surface antigen (HBsAg) not being present and low DNA viral load in serum. In this study, an attempt was made to investigate the outbreak of anti-HBc and OBI among the HBsAg-negative donors in Golestan province. Materials and Metho...

Prevalence of "anti-HBc alone" among Syrian blood donors.

INTRODUCTION We aimed to evaluate the prevalence of "anti-HBc alone" among Syrian blood donors, highlighting the possibility of representing occult HBV infection. METHODOLOGY Sera of 3,896 healthy blood donors were tested for both HBsAg and anti-HBc. HBsAg-negative, anti-HBc-positive samples were further tested for the antibodies to HBsAg (anti-HBs), and "anti-HBc alone" sera were tested for ...

Serological evidence of presence of HBsAg undetectable by conventional radioimmunoassay in anti-HBc positive blood donors.

Sera from two blood donors, one of whom was implicated in a case of post-transfusion hepatitis B, were found to be positive for anti-HBc and negative for HBsAg by conventional radioimmunoassay and were retested for HBsAg after concentration (pepsin digestion and polyethylene glycol precipitation). The presence of occult HBsAg was confirmed in both. These observations have implications for blood...

Prevalence of HBc-Ab among HBs-Ag negative healthy blood donors in south of Iran

BACKGROUND The risk of infection with transfusion-transmitted viruses, such as Hepatitis B virus (HBV), has been reduced markedly. However, a zero-risk blood supply remains a popular goal. Anti-hepatitis B core antigen (anti-HBc) is considered to be an effective marker for occult HBV infection, and it has served a significant role in improving blood safety. The purpose of this study was to dete...

Indication of Anti-HBc Antibody Screening and HBV-DNA Detection in Diagnosing Latent Hepatitis B Virus Infection

Background: In spite of available, and sensitive screening assay for detection of hepatitis B virus surface antigen (HBsAg), occasional cases of post-transfusion hepatitis B virus infection are still observed. The aim of the present study was to assess the prevalence of positive anti hepatitis B core (anti-HBc) and presence of HBV-DNA in serum sample of healthy blood donors negative for both HB...
