چکیده مقاله:

Objective(s): The purpose of this study is to investigate the indication function of the calcium circulation-related factors on the damage to muscle strength and contraction function after nerve injury. The target factors include ryanodine receptor (RyR), inositol-1,4,5-triphosphate receptor (IP3R), phospholamban (PLN), cryptocalcitonin (CASQ), ATPase and troponin C (TNNC).Materials and Methods: Sprague-Dawley (SD) rats were randomly divided into sham-operated group (SO), sciatic nerve injury group (SNI) and sciatic nerve disconnection group (SNT). Sciatic nerve function index and stretching test were used to examine the changes to muscle strength; bilateral gastrocnemius muscles were extracted after execution for gastrocnemius wet weight ratio test. HE staining slides and average cross-sectional area of muscle fibers were acquired to analyze the muscle atrophy. The transcription level of the factors was also measured. Results: Sciatic nerve damage in SNI group was significantly higher than that in SO group in the 6 weeks, but there was no significant difference between SNT and SO groups fallowing sciatic nerve damage. Sciatic nerve function in SNT group was worse than that in SNI group. The average cross-sectional area of gastrocnemius muscle fibers in SNI and SNT groups was significantly reduced compared to that in SO group. The transcriptional levels of RyR, PLN, CASQ, ATPase and TNNC in SNI and SNT groups were significantly different from those in SO group. Conclusion: Calcium circulation-related factors could be used as potential indicators for assessment of damages to muscle strength.

Misdirection of regenerating motor axons after nerve injury and repair in the rat sciatic nerve model.

Misdirection of regenerating axons is one of the factors that can explain the poor results often found after nerve injury and repair. In this study, we quantified the degree of misdirection and the effect on recovery of function after different types of nerve injury and repair in the rat sciatic nerve model; crush injury, direct coaptation, and autograft repair. Sequential tracing with retrogra...

Age-Dependent Regeneration by Using Electromyographical Study Foliowing Sciatic Nerve Injury in Rat

Purpose: There are extensive evidences that show axonal processes of the nervous system (peripheral and/or central) may be degenerated after nerve injuries. Axonal regeneration is relation to various factors. In this investigation we decided to evaluate the effects of nerve regeneration age-dependent on injured rat sciatic nerv. Materials and Methods: For this study, the right sciatic nerve of...

Dexmedetomidine to Help Nerve Regeneration in a Rat Sciatic Nerve Injury Model

Background Several studies have shown that dexmedetomidine (DXM), a selective α2-adrenoceptor agonist, also has neuroprotective effects. However, its effect on impaired peripheral nerve regeneration has not been studied. Materials and Methods Forty-five Sprague-Dawley rats were randomly assigned to three groups: group 1 (control SHAM), group 2 (sciatic nerve injury + normal saline), and group...

A Rare Anatomical Variation of Sciatic Nerve and its Relationship with Piriformis Muscle

Sciatic nerve is the thickest nerve of the body which is formed by ventral branches of L4, L5, S1, S2 and S3 of spinal nerves as a part of sacral plexus in pelvis. It leaves the pelvis via the greater sciatic foramen below piriformis muscle and descends between the greater trochanter and ischial tuberosity, along the back of the thigh and dividing into the tibial and common peroneal (fibular) n...

P49: The Evaluation of Aqueous Extract of Glycyrrhiza Glabra on Nerve Recovery in the Rat after Sciatic Nerve Injury

Peripheral nerve injury requires a long recovery period, and recovery, once attained, usually is incomplete. Inflammatory procedures may inhibit functional recovery after nerve injury and produce cell death in both the central nervous system and the peripheral nervous system. Since the glycyrrhiza glabra extract has anti-inflammatory effects, it could reduce the severity of injury. The aim of t...
