Genistein and bromotetramisole (Br-t) strongly activate cystic fibrosis transmembrane conductance regulator (CFTR; ABCC7) chloride channels on Chinese hamster ovary cells and human airway epithelial cells. We have examined the possible role of phosphatases in stimulation by these drugs using patch-clamp and biochemical methods. Genistein inhibited the spontaneous rundown of channel activity tha...

1. Some properties of the triphosphoinositide phosphomonoesterase from the homogenates of guinea-pig brain were studied. The enzyme has an optimum pH range 6.7-7.3, is stimulated with KCl at a concentration of 0.1M, and under these conditions has K. 1.43 x 10-4M. 2. A factor from the 'pH5 supernatant' of guinea-pig brain stimulates the enzyme activity over and above the stimulation produced by ...
In the present study we have localized acid and alkaline phosphatases, glucose-6-phosphatase and sodium-potassium ATPase (Na-K-ATPase) in ultra-thin sections of cerebral neurosecretory cells and oesophageal cells of the earthworm, Aporrectodea caliginosa. The majority of the enzymes studied were located in the rough endoplasmic reticulum (RER), nuclear envelope and Golgi apparatus.
The alkaline phosphatases of 29 strains of bacteria assigned by various authors to the genera Aerobacter, Klebsiella and Enterobacter were compared by the micro-complement fixation technique. On the basis of phosphatase resemblance, we recommend that all strains hitherto assigned to Aerobacter aerogenes and Enterobacter aerogenes be assigned to the genus Klebsiella.
The evolution of the alkaline phosphatase (AP) gene family has lead to the existence in humans of one tissue-nonspecific (TNAP) and three tissue-specific isozymes, i.e. intestinal (IAP), germ cell (GCAP), and placental AP (PLAP). To define the structural differences between these isozymes, we have built models of the TNAP, IAP, and GCAP molecules based on the 1.8-structure of PLAP(1) and have p...
The two main alkaline phosphatase isoenzymes in serum from patients with extrahepatic biliary obstruction have been purified. Properties of these isoenzymes, such as electrophoretic mobility, separation on gel filtration, ultracentrifugation characteristics, Michaelis constants, and sensitivity to neuraminidase have been studied and compared with the isoenzymes of liver and bile. The results sh...
Two extracellular, heat-labile alkaline phosphatases were purified from a psychrophilic Arthrobacter isolate, D10. The enzymes were active over different pH ranges, used distinct substrates, and had different kinetic properties. Each enzyme reacted specifically to its own antibody during immunoblot analysis. One had both monophosphatase and diesterase activities.
This report relates to the preceding paper by Taniuchi and Anfinsen. Micrococcal nuclease from the strain “Foggi” has been purified from a commercially available source (Worthington). The enzyme was crystallized from an ammonium sulfate solution. The over-all purification was approximately SOO-fold with an over-all yield of about 16%. The enzyme is virtually devoid of acid and alkaline phosphat...
Peptides with different chromatographic and electrophoretic properties were obtained from human placental and renal alkaline phosphatases by tryptic digestion of the enzymes labelled with radioactive orthophosphate at their active centres. These results provide structural evidence for the distinct genetic origins of the two isoenzymes that had previously been inferred from their different prope...
