Hematopoiesis is defined as the ongoing production of blood cells. As most mature blood cells are relatively short-lived and require continuous replacement, hematopoiesis is characterized by an extraordinary turnover rate with production of trillions of new blood cells every day. To cope with the enormous proliferation required to generate sufficient numbers of blood cells to maintain homeostas...
We previously reported a transgenic mouse model expressing herpesvirus thymidine kinase (TK) gene under the control of a 2.3-kilobase fragment of the rat collagen alpha1 type I promoter (Col2.3 Delta TK). This construct confers lineage-specific expression in developing osteoblasts, allowing the conditional ablation of osteoblast lineage after treatment with ganciclovir (GCV). After GCV treatmen...
Homozygous loss of function of Runx1 (Runt-related transcription factor 1 gene) during murine development results in an embryonic lethal phenotype characterized by a complete lack of definitive hematopoiesis. In light of recent reports of disparate requirements for hematopoietic transcription factors during development as opposed to adult hematopoiesis, we used a conditional gene-targeting stra...
Background: Protection of hematopoietic system has become a primary goal in the development of novel medical countermeasures against ionization radiation and radiotherapy. This study was to explore the role of rapamycin in normal tissues against radiation. Materials and Methods: Mice were pretreated with rapamycin by i.p. every other day for five times before 5 Gy or 8.5 Gy γ-ray whole bo...
Hematopoiesis is a biological phenomenon (process) of production mature blood cells by cellular differentiation. It based on amplification steps due to an interplay between renewal and differentiation in the successive cell types from stem cells. We will study this mechanism with stochastic point view explain unexpected fluctuations number, as surprisingly observed biologists medical doctors re...
Hematopoiesis has been studied via stem cell labeling using barcodes, viral integration sites (VISs), or in situ methods. Subsequent proliferation and differentiation preserve the tag identity, thus defining a clone of mature cells across multiple type lineages. By tracking population clones, measured within samples taken at discrete time points, we infer physiological parameters associated wit...
We report a case of intrathoracic extramedullary hematopoiesis associated with hemolytic anemia. While the paravertebral localization of the lesions demonstrated on CT and MRI was suspicious of intrathoracic extramedullary hematopoiesis, In-111 chloride and Tc-99m Sn colloid bone marrow scintigraphies showing a bone marrow element of the lesion were useful to confirm the diagnosis.
The transcription factors SCL/Tal-1 and AML1/Runx1 control the generation of pluripotent hematopoietic stem cells (pHSC) and, thereby, primitive and definitive hematopoiesis, during embryonic development of the mouse from mesoderm. Thus, Runx1-deficient mice generate primitive, but not definitive hematopoiesis, while Tal-1-deficient mice are completely defective. Primitive as well as definitive...
anemia is the most common hematologic disorder in patients with ulcerative colitis (uc). in some cases, normochromic anemia results from the presence of chronic disease; however blood loss or malabsorption may lead to an iron deficiency anemia with hypochromic appearance. other rare hematologic manifestations associated with uc include myelodysplastic syndromes and leukemia. several investigato...
Maintenance of cell pluripotency, differentiation, and reprogramming are regulated by complex gene regulatory networks (GRNs) including monoallelically-expressed imprinted genes. Besides transcriptional control, epigenetic modifications and microRNAs contribute to cellular differentiation. As a model system for studying the capacity of cells to preserve their pluripotency state and the onset of...
