神経変性疾患の多くは、アンメット・メディカル・ニーズの高い疾患である。その中で、アルツハイマー病、進行性核上性麻痺や前頭側頭葉変性症などのタウオパチーは、タウ蛋白が異常なリン化により細胞内にタウ凝集体として蓄積し、さらにこの凝集体が神経細胞を伝播して、その結果、神経変性が生じる。仮に神経細胞内でタウ凝集体の蓄積を抑制する薬物が見出されたとしても、その薬物を効率よく神経細胞内に送達でき、しかも神経細胞を乗り継いで移行できるDDS技術が開発されなければ、神経変性疾患の進行を阻止し、根本的に治療する有効な治療薬を開発することは難しい。そこで本稿では、筆者が、神経変性疾患に適用可能で、神経ペプチドを神経細胞内の軸索を介して神経細胞を乗り継いで側脳室内投与よりも効率よく作用部位へ送達させ、中枢作用を示すことのできるNose-to-Brainシステムを開発したので、紹介する。
