Here we report the development of a gene-synthesis technology, circular assembly amplification. In this approach, we first constructed exonuclease-resistant circular DNA via simultaneous ligation of oligonucleotides. Exonucleaseand subsequent mismatch cleaving endonuclease–mediated degradation of the resulting ligation mixture eliminated error-rich products, thereby substantially improving gene...

A DNA machine finds its application to construct an enzyme-free, label-free, cost-effective and novel approach for amplified DNA detection. The present approach is endowed with high sensitivity and selectivity. A detection limit as low as 0.2 nM was obtained by practical measurement.

In this paper, we construct linear codes over Z4 with bounded GCcontent. The codes are obtained using a greedy algorithm over Z4. Further, upper and lower bounds are derived for the maximum size of DNA codes of length n with constant GC-content w and edit distance d. keywords: DNA codes, GC-content, edit distance, upper and lower bounds.

This protocol describes how to transform moss (Physcomitrella patens) protoplasts using polyethylene glycol (PEG)-mediated DNA uptake. The transformation rates for direct uptake by protoplasts of DNA with and without genomic sequence (a targeting construct) are typically 10 and 10, respectively. (These are the frequencies of stable transformants among regenerants surviving the transformation pr...

We describe here an improved method for isolating, purifying, and cloning DNA from diverse soil microbiota. Soil microorganisms were extracted from soils and embedded and lysed within an agarose plug. Nucleases that copurified with the metagenomic DNA were removed by incubating plugs with a high-salt and -formamide solution. This method was used to construct large-insert soil metagenomic librar...

Allosteric control of deoxyribozymes is useful for a broad range of practical applications, such as nucleic acid sensing and DNA-computing. We found that the catalytic activity of a DNA-hydrolyzing deoxyribozyme could be allosterically regulated by adding short oligonucleotides. We used this technique to construct deoxyribozyme-based logic gates.

Oxidative stress may play an important role in the carcinogenic action of UVB light (290 to 320 nm). UVB light induces the growth-related immediate-early gene c-fos in JB6 mouse epidermal cells, but at the same time it causes structural damage to DNA, in particular DNA strand breakage. We have studied the effect of the modulation of the frequencies of DNA breaks on the transcriptional induction...

We have developed a method for introducing mutations into the adenovirus type 2 genome at predetermined sites. Specific mutations are introduced into segments of the viral genome cloned in bacteria by using plasmid vectors. The chimeric DNA is used to construct viral mutants by cotransfection with two viral DNA segments derived from both ends of the viral genome, each of which has overlapping s...

Due to the complexity and heterogeneity of cancer, the development of cancer diagnosis and therapy is still progressing, and a complete understanding of cancer biology remains elusive. Recently, cancer nanomedicine has gained much interest as a promising diagnostic and therapeutic strategy, as a wide range of nanomaterials possess unique physical properties that can render drug delivery systems...
