The purification of the alkaline phosphatases of cow's milk and calf intestinal mucosa as previously described (Morton, 1950, 1953a, 1954a) was initiated in order to enable investigation of the properties of these enzymes. The need for such a study has been well recognized (see Moog, 1946, for example), but hitherto all work has been carried out with crude or partially purified enzyme preparati...

Two alkaline phosphatases were extracted from the membranes of Bacillus subtilis 168 stationary-phase cells and purified as homogeneous proteins by hydroxyapatite column chromatography. Alkaline phosphatases I and II differed in several properties such as subunit molecular weight, substrate specificity, thermostability, Km, pH stability, and peptide maps.

This paper reviews the alkaline phosphatases in canine serum, the analytical methods used for qualitative and/or quantitative detection of these isoenzymes, and the diagnostic significancy of each of these isoenzymes. The paper further describes some of the latest advances in our knowledge of the canine alkaline phosphatases and possible areas of future research.

The immunochemical differentiation of the alkaline phosphatases from tissues of the dog has been reported (2, 3). Antisera against dog intestinal phosphatase quantitatively precipitated this enzyme under conditions which were without effect on the alkaline phosphatases from liver and kidney of the same species. The potential value of this immunochemical approach for studies of tissue specificit...

Approximately 10% of the alkaline phosphatase activity in human kidney is derived from the intestinal-type alkaline phosphatase isoform, which can be differentiated from adult intestinal alkaline phosphatase by selective reactivity with monoclonal antibodies. The NH2-terminal sequence of the renal intestinal-type alkaline phosphatase was shown to be identical to sequences of the adult and mecon...

Microorganisms producing alkaline phosphatases are wide spread in nature. Alkaline phosphatase (Orthophosphoric monoester phosphohydrolase, E.C.3.1.3.1) is a hydrolase enzyme functioning at alkaline pH. The metalloenzyme containing the two zinc ions are involved in catalysis whereas the magnesium ion is important in structural stabilization (Simpson et al., 1968; Anderson et al., 1975). The enz...

Two enzyme forms of alkaline phosphatase have been partially purified from the medium spent for the culture of HUH-6 clone 5 cells, which were originally derived from hepatoblastoma tissue. The purification methods used are ammonium sulfate precipitation, ethanol precipitation, diethylaminoethyl cellulose chromatography, Affi-Gel Blue chromatography, and Sephadex G-200 gel filtration. These alk...

Alkaline phasphatase occurs in several tissues and consequently the source of the enzyme in serum has stimulated considerable investigation and speculation. Several workers (l-3) have studied the effects of diet upon intestinal and serum alkaline phosphatase activity, and, in general, similar dietary regimens have similar effects on the two enzymes. The serum enzyme shows a significant correlat...
