5-Lipoxygenase-derived products have been implicated in both the inhibition and promotion of chronic infection. Here, we sought to investigate the roles of endogenous 5-lipoxygenase products and exogenous leukotrienes during Histoplasma capsulatum infection in vivo and in vitro. 5-LO deficiency led to increased lung CFU, decreased nitric oxide production and a deficient primary immune response ...
Histoplasmosis infection is endemic in Asia and disseminated histoplasmosis (DH) is one form of its presentation (Benevides et al., 2007[1]). DH commonly affects both adrenal glands. We describe a case of disseminated histoplasmosis complicated with hypercalcaemia in a 75-year-old immunosuppressed patient who presented with bilateral adrenal masses. The fine needle aspiration cytology of the ad...
A medium is described for the maintenance of the mycelial phases of Histoplasma capsulatum in their "original" condition of sporulation. A yeast phase medium is reemphasized, not only for conversion and support of the yeast phase, but as an indirect method of maintaining the mycelial characteristics as originally observed.
Monoclonal antibodies to Histoplasma capsulatum can modify pathogenesis. We now show that monoclonal antibody H1C to a 70-kDa antigen increases intracellular fungal growth and reduces macrophage nitric oxide release but has no effect on fungal burden or survival in murine infection. This further demonstrates the complexities of host-pathogen interactions.
The prevalence of fungal infections has increased in immunocompromised patients, leading to millions deaths annually. Arachidonic acid (AA) metabolites, such as eicosanoids, play important roles regulating innate and adaptative immune function, particularly since they can function virulence factors enhancing colonization are produced by mammalian lower eukaryotes, yeasts other fungi (Candida al...
The YPS3 gene of Histoplasma capsulatum encodes a protein that is both surface localized in the cell wall of H. capsulatum and released into the culture medium. This protein is produced only during the pathogenic yeast phase of infection and is also expressed differentially in H. capsulatum strains of different virulence levels. In this study, we silenced the YPS3 transcript by using an interfe...
