Here, we developed an isogenic cell model of "stemness" to facilitate protein biomarker discovery in breast cancer. For this purpose, we used knowledge gained previously from the study of the mouse mammary tumor virus (MMTV). MMTV initiates mammary tumorigenesis in mice by promoter insertion adjacent to two main integration sites, namely Int-1 (Wnt1) and Int-2 (Fgf3), which ultimately activates...

Cellular resistance to the antifolate methotrexate (MTX) is often caused by target amplification, uptake defects, or alterations in polyglutamylation. Here we have examined MTX cross-resistance in a human breast carcinoma cell line (MCF7/MX) selected in the presence of mitoxantrone, an anticancer agent associated with the multidrug resistance (MDR) phenotype. Examination of protein expression a...

Accumulating evidence indicates that breast cancer is caused by cancer stem cells and cure of breast cancer requires eradication of breast cancer stem cells. Previous studies with leukemia stem cells have shown that NF-jB pathway is important for leukemia stem cell survival. In this study, by using MCF7 sphere cells as model of breast cancer stem-like cells, we evaluated the effect of NF-jB pat...

Background and purpose: Chalcones are promising lead for anticancer drug design and discovery. Chalcones with different mechanisms including tubulin polymerization inhibition induce apoptosis in cancer cells. The aim of current work was synthesis of two indole-chalcone derivatives and investigation of their cytotoxic activity against cancer and normal cell lines, as well as molecular docking st...

MCF7 human breast cancer cells selected for resistance to doxorubicin (adriamycin; DoxR) have developed the phenotype of multidrug resistance. Multidrug resistance in DoxR MCF7 cells (called AdrR MCF7 cell line in previous publications) is associated with biochemical changes similar to those induced by carcinogens in rat hyperplastic liver nodules (HNs) and associated with resistance to xenobio...

In cancer, mucins are aberrantly O-glycosylated, and consequently, they express tumor-associated antigens such as the Tn determinant (alpha-GalNAc-O-Ser/Thr). As compared with normal tissues, they also exhibit a different pattern of expression. In particular, MUC6, which is normally expressed only in gastric tissues, has been detected in intestinal, pulmonary, colorectal, and breast carcinomas....

The p21 belongs to the CIP/KIP family of CDK inhibitors involved in cell cycle arrest at specific stages of the cell cycle progression. DNA methylation is the best studied epigenetic mark that have been evidently associated to chromatin condensation, and repression of gene transcription. The CpG island hypermethylation in promoter region of certain genes occurs in cancer cells and affects tumor...

Although the epithelial-mesenchymal transition (EMT) is a normal process that occurs during development, it is thought to be associated with cancer progression and metastasis. Emerging evidence links mesenchymal stem cells (MSCs) in the tumor microenvironment with the occurrence of EMT in cancer progression. In this study, the human breast cancer cell line MCF7 was co-cultured with human adipos...

a series of fluoroquinolone derivatives holding triazolidindione moieties have been synthesized and proved to be cytotoxic agents in vitro particularly against cancer cell lines (sknmc, mcf7, a2780-cp, sw48, a549, kb, ht-29, hepg 2 ). the cytotoxic activity was assessed using mtt colorimetric assay. our compounds had less cytotoxicity than doxorubicin in all studied cell lines.
