A short report has recently been made (1) concerning the preparation of an acetyl derivative of chymotrypsin containing a single acetyl group of relatively high reactivity. The reaction, which may be carried out by any of several nitrophenyl acetates, begins with a rapid initial liberation of the free nitrophenol in practically mole for mole proportions to the total chymotrypsin present. Therea...

Rhoads, M. L., Fetterer, R. H., Hill, D. E., and Urban, J. F., Jr. 2000. Trichuris suis: A secretory chymotrypsin/elastase inhibitor with potential as an immunomodulator. Experimental Parasitology 95, 36– 44. A serine protease inhibitor, termed TsCEI, was purified from adultstage Trichuris suis by acid precipitation, affinity chromatography (elastase–agarose), and reverse-phase HPLC. The molecu...

The pancreatic function diagnostant (PFD test) and fecal chymotrypsin test were performed simultaneously in patients after pancreatic surgery, and findings for both tests were analyzed. The paminobenzoic acid recovery rate in the PFD test and the chymotrypsin activity in stools decreased after pancreatectomy, particularly after extended radical pancreaticoduodenectomy. Chymotrypsin activity in ...

Mast cells contain an enzyme which hydrolyzes 3-chloroacetoxy-2-naphthoic acid anilide. By using highly purified mast cells isolated by differential centrifugation in high density sucrose solutions we have been able to study this enzymatic activity in more detail. The enzyme has properties similar to those of chymotrypsin: Chymotrypsin will hydrolyze the histochemical substrate, and the chymotr...

The geometry of the binary and ternary complexes of two black-eyed pea inhibitors with trypsin and chymotrypsin has been established by distance measurements using the technique of singlet-singlet energy transfer. Triangulation of measured distances in the ternary double-headed complex of the trypsin-chymotrypsin inhibitor (BEPCI) with trypsin and chymotrypsin limits the possible structural mod...

PURPOSE Cytogenetic abnormalities are currently the most important predictors of response and clinical outcome for patients with acute myeloid leukemia (AML) or advanced-stage myelodysplastic syndrome (MDS). Because clinical outcomes vary markedly within cytogenetic subgroups, additional biological markers are needed for risk stratification. EXPERIMENTAL DESIGN We assessed the utility of meas...

We describe a novel 'switch-off' biosensing strategy for the detection of chymotrypsin based on surface plasmon resonance (SPR) and surface-enhanced Raman scattering (SERS) spectroscopy. This approach analyzes the fingerprint spectrum of the chymotrypsin-catalyzed substrate. The lowest probed concentration is 0.4 nM for chymotrypsin.

For several years this laboratory has been concerned with an investigation of those enzymes in rat skin extracts which, in part at least, comprise the proteolytic enzyme system of this organ (l-5). In certain cases, methods have been proposed for the detection of their individual activities. However, certain contemplated studies, e.g. the possible physiological significance of these proteolytic...

Previous experimental and theoretical studies have produced high-resolution descriptions of the native and folding transition states of chymotrypsin inhibitor 2 (CI2). In similar fashion, here we use a combination of NMR experiments and molecular dynamics simulations to examine the conformations populated by CI2 in the denatured state. The denatured state is highly unfolded, but there is some r...

Peptide mapping is routinely used in the characterization of monoclonal antibodies (mAbs) for confirmation primary sequence and detection post-translational modifications (PTMs). Trypsin one most commonly proteases peptide protocols due to its high level specificity. However, it has been observed that trypsin alone not always sufficient full coverage because presence long sequences hydrophobic ...
