One of the specific inhibitors of calmodulin action, trifluoperazine, blocked the stimulating action of insulin on 2-deoxyglucose uptake and glucose metabolism. The inhibitory effect of insulin on lipolysis was not altered by the drug. The active (insulin-stimulated) state and the basal state of lipogenesis were inhibited half-maximally at 80 and 550 microM trifluoperazine, respectively. 2-Deox...
Conversion of phosphorylase b to a which is catalyzed by the enzyme phosphorylase kinase is known to require Ca++. Trifluoperazine, an inhibitor of calmodulin-dependent enzymes, was utilized in the present study to clarify the role in vivo of calcium-calmodulin regulation of phosphorylase kinase. Twenty-minute preperfusion of isolated rat ventricles with 10(-5) M trifluoperazine had no effect o...
A 22-year-old man presented to the casualty department with "locking" of his right great toe in dorsiflexion, and of his jaw. One week previously, he had complained of nausea and a sore throat for which his general practitioner had prescribed a course of oral penicillin. He also suffered from intermittent pain arising from a wisdom tooth, and he had been involved in a fight four days previously...
Liver injury occurs with many drugs; therefore, a thorough work up is important for establishing the diagnosis. We report a case of trifluoperazine-induced cholestatic jaundice. A 44-year old male with schizoaffective disorder developed an increase in liver enzymes and jaundice after starting treatment with trifluoperazine. Workup for other potential etiologies was negative.
A B S T R A C T Cultured, spontaneously beating heart cells were used to study the role of calmodulin in regulating cardiac contraction. Trifluoperazine (TFP), an antipsychotic drug that binds to calmodulin, reversibly inhibited myocardial contraction. This effect occurred over a TFP concentration range of 5-100 MM with half maximal activity at -15 uM TFP. When the phosphoprotein profile of TFP...
This case report outlines a rare presentation of hypokalemia with therapeutic doses of trifluoperazine in a patient with schizophrenia. Although there was comorbid diabetes mellitus in the patient, its effect was ruled out subsequently. On recovery, rechallenge with trifluoperazine resulted in the same effect at a milder level. No cardiac abnormalities were detected with the presentation. Naran...
Using immuno electron microscopy we have identified a number of calmodulin/calmodulin binding proteins distributed at various subcellular sites (Eastlake et al, 1994). Treatment of H. diminuta with trifluoperazine has potential cestocidal properties influencing both tegumental protein release, activity of calmodulindependent enzymes and the uptake of glucose (Branford White & Hipkiss, 1985; Hip...
Resistance to the cytotoxic effects of daunomycin and Adriamycin (ADR) in sublines of Ehrlich ascites and P388 mouse tumors has been demonstrated to be due to reduced cellular accumulation and retention of drug. In this study, the effect of the calmodulin inhibitor trifluoperazine on the cellular accumulation, retention, and cytotoxic effects of ADR in ADR-sensitive (P388/S) and ADR-resistant (...
A 29-year-old man admitted to a psychiatric unit was successfully treated for a paranoid psychosis with trifluoperazine. Following reduction of neuroleptic dosage and addition of paroxetine he developed severe extra-pyramidal side effects. These remitted after withdrawal of medication and did not recur following recommencement of trifluoperazine. This is the first report of such a side effect a...
