OBJECTIVES To determine whether the fatty acid synthesis enzyme malonyl coenzyme A:acyl carrier protein transacylase (MCAT) is involved in the growth-inhibitory effect of trifluoperazine in the tubercle bacillus Mycobacterium bovis BCG. METHODS BCG was grown in liquid culture with various concentrations of trifluoperazine and growth was monitored by OD measurement. To determine the effect of ...

The effects of trifluoperazine on the activation of glycogenolysis by various hormones were studied in perfused rat liver. Trifluoperazine significantly inhibited glycogenolytic effect of phenylephrine and angiotensin II by lowering maximal response, and that of vasopressin by shifting the dose-response curve to the right, while alpha-antagonist phentolamine was inhibitory only to phenylephrine...

Trifluoperazine, a commonly used antipsychotic drug, has been known to induce QT prolongation and torsades de pointes, which can cause sudden death. We studied the effects of trifluoperazine on the human ether-a-go-go-related gene (HERG) channel expressed in Xenopus oocytes and on the delayed rectifier K(+) current of guinea pig cardiomyocytes. The application of trifluoperazine showed a dose-d...

A B S T R A C T The role of calmodulin in insulin secretion from rat pancreatic islets has been examined by the use of trifluoperazine, an inhibitor of calmodulin-Ca++-directed functions. It was found that 30 ,M trifluoperazine caused 50% inhibition, and 100 ,M, up to 73% inhibition of 16.7 mM glucosestimulated insulin release. 100 ,M trifluoperazine caused a similar inhibition of 10 mM glycera...

Angioedema is a serious adverse drug reaction that can rarely be associated with trifluoperazine treatment. We present the case of a 44-year-old male with an established diagnosis of schizoaffective disorder, for which trifluoperazine therapy was considered. He presented to the emergency department with bilateral lower limb oedematous painful erythematous swelling that eased off completely when...

Calsequestrin is an intermediate affinity, high capacity Ca(2+)-binding protein found in the lumen of the sarcoplasmic reticulum of both skeletal and cardiac muscle cells. Previous sequence analysis suggested that calsequestrin may contain a hydrophobic binding site for the drug trifluoperazine, a site shared by the calmodulin family and shown to play a role in calmodulin/calmodulin receptor in...

The calmodulin antagonist trifluoperazine inhibits many responses of human platelets to stimuli such as thrombin. We have now demonstrated that trifluoperazine inhibits the production of all arachidonic acid metabolites, by both cyclooxygenase and lipoxygenase pathways, in thrombin-stimulated platelets. The metabolism of exogenously supplied arachidonlc acid was not affected by trifluoperazine....

The possible involvement of calmodulin in insulin release was evaluated by studying the effects on intact islets of trifluoperazine and pimozide, two antipsychotic agents known to bind strongly to calmodulin in cell-free systems. Trifluoperazine (10-100#UM) produced a doseand time-dependent inhibition of the two phases of glucose-stimulated insulin release. The effect was not reversible by simp...

liver injury occurs with many drugs; therefore, a thorough work up is important for establishing the diagnosis. we report a case of trifluoperazine-induced cholestatic jaundice. a 44-year old male with schizoaffective disorder developed an increase in liver enzymes and jaundice after starting treatment with trifluoperazine. workup for other potential etiologies was negative.
