During vertebrate embryogenesis, the primary body axis grows posteriorly and is concomitantly segmented into somites, the precursors of the vertebral column, skeletal muscle, and dermis. The somites arise sequentially, with the anterior somites that give rise to the cervical vertebrae created early. The more posterior somites that become the thoracic, lumbar, and sacral vertebrae form at progre...

Somite development is governed tightly by genetic factors. In the large-scale mutagenesis screens of zebrafish, no mutations were linked to myocyte enhancer factor 2A (MEF2A) locus. In this study, we find that MEF2A knock-down embryos display a downward tail curvature and have U-shaped posterior somites. Furthermore, we demonstrate that MEF2A is required for Hedgehog signaling. MEF2A inhibition...

Somites are the bilaterally symmetric embryonic precursors of the vertebrate skeleton and axial muscle. Three recent studies reveal that somites form asymmetrically in the absence of retinoic acid signaling. These results uncover an unexpected relationship between somitogenesis and left-right patterning, and suggest that bilateral somite formation is regulated along the left-right axis.

We have developed a whole-mount immunocytochemical method for Xenopus and used it to map the expression of the intermediate filament protein vimentin during early embryogenesis. We used two monoclonal antibodies, 14h7 and RV202. Both label vimentin filaments in Xenopus A6 cells, RV202 reacts specifically with vimentin (Mr, 55 x 10(3] on Western blots of A6 cells and embryos. 14h7 reacts with vi...

The migratory pathways of neural crest-derived pigment cells were examined in two anurans, Xenopus laevis and Discoglossus pictus, and correlated with the distribution of glycosaminoglycans (GAG) in the extracellular matrix (ECM) of these pathways. In Xenopus, melanophores in the trunk reach the dermis by initially migrating ventrally, between the neural tube and somites, and then by migrating ...

Signals originating from tissues surrounding somites are involved in mediolateral and dorsoventral patterning of somites and in the differentiation of the myotome. Wnt-1 and Wnt-3a, which encode members of the Wnt family of cystein-rich secreted signaling molecules, are coexpressed at the dorsal midline of the developing neural tube, an area adjacent to the dorsomedial portion of the somite. Se...

The regional embryonic origin of trunk and limb musculature was determined through heterospecific homotopic or heterotopic transplantations of quail somitic or somiticsomatopleural mesoderm into chick hosts, and through localized X-irradiation of the somitic mesoderm. Experiments were performed on 2-day embryos. Results show that the myoblastic component of all truncal and appendicular muscles ...
