Over 100 patients with lepromatous leprosy were treated with rifampicin in a series of pilot, uncontrolled, and controlled trials in 1968-77. The rapid bactericidal effect of rifampicin on Mycobacterium leprae was confirmed. Clinical improvement became apparent sometimes as early as 14 days after the start of treatment. Nevertheless, a few persisting viable M leprae were detected as long as fiv...
INTRODUCTION Multidrug therapy for leprosy is currently done with dapsone, clofazimine and rifampicin. Dapsone is known to cause hemolytic anemia (HA) and this adverse event during MDT seems to be more frequent than reported. The aim of this report is to discuss and grade HA due to dapsone during MDT treatment for leprosy. METHODS This is a retrospective study of 194 leprosy patients from a L...
The in vitro activity of trimethoprim plus dapsone against oxacillin-resistant Staphylococcus aureus was comparable to that of trimethoprim plus sulfamethoxazole. Because of its different pharmacologic properties, dapsone may be a useful agent in combination with trimethoprim for the treatment of patients colonized with oxacillin-resistant S. aureus.
Molecular detection was compared with the mouse footpad inoculation test for detection of dapsone resistance in 38 strains of Mycobacterium leprae. Mutations of the folP1 gene (at codons 53 or 55) were found in 6 of 6 strains with high-level resistance, in 3 of 4 strains with intermediate-level resistance, and in 1 of 6 strains with low-level resistance, but not in 22 dapsone-susceptible strain...
Dapsone has been used successfully as adjuvant therapy for bullous pemphigoid (BP). The effectiveness of dapsone for this indication, however, remains controversial. We evaluated the effectiveness and adverse events of dapsone (1.0-1.5 mg/kg per day) in combination with oral methylprednisolone (tapering doses of 0.5 mg/kg per day) and topical clobetasol propionate (initially applied once daily ...
U n der the conditions of this s tudy, dapsone in d oses of 5 mg and 10 mg daily when ad ministered to p atients with lepromat ous leprosy is an ineffe ctive therapy in terms of killing and elimin ation of My co. leprae from human skin and b one marrow . A real danger of facili tating the emergence of resistant bacilli exists . Therefore , un til there is more evidence from long term therapeuti...
OBJECTIVE To Study the incidence and clinical characteristics of dapsone hypersensitivity syndrome (DHS) among MDT-treated leprosy patients from 2006 to 2009 in China. METHODS A retrospective survey was carried out throughout China using a specially designed questionnaire. RESULTS From 2006 to 2009, there were 63 new patients reported to have DHS with an incidence of 1.0%. Among these patie...
Three patients who developed hypersensitivity reaction at varying intervals after the initiation of dapsone therapy are described. Two of them had leprosy whereas the other had dermatitis herpetiformis. Their clinical manifes tations were not uniform. Although fatal complications have been reported, the unique role played by dapsone in the treatment of leprosy and the importance of recognizing...
Pyrimethamine-sulfadoxine, the first choice for uncomplicated falciparum malaria in Africa, exerts strong selection pressure for resistance because of its slow elimination. It is likely that resistance will emerge rapidly, and there is no widely affordable replacement. Chlorproguanil-dapsone is cheap, rapidly eliminated, more potent than pyrimethamine-sulfadoxine, and could be introduced in the...
