The synthesis of a library of nucleoside triphosphate mimetics is described where the Mg(2+) chelated triphosphate sidechain is replaced by an uncharged methylene-triazole linked monosaccharide sidechain. The compounds have been evaluated as inhibitors of Bacillus anthracis pantothenate kinase and a competitive inhibitor has been identified with a K(i) that is 3-fold lower than the K(m) value o...
Pantothenate kinase (CoaA) catalyzes the first step of the coenzyme A biosynthetic pathway. Here we report the identification of the Staphylococcus aureus coaA gene and characterization of the enzyme. We have also identified a series of low-molecular-weight compounds which are effective inhibitors of S. aureus CoaA.
fluid, and Tables 3 and 6 that the glycolysis of suspensions is unaffected by the compound. A small stimulation only was fotmd in previous experiments with the same organism (McIlwain, 1944) in which, in contrast with the present work, the course of glycolysis was measured. Pantothenate is always present in small quantities in the organisms; nevertheless, if free pantothenate is present, it is ...
Toennies, G. (Temple University School of Medicine, Philadelphia, Pa.), D. N. Das, and F. Feng. Pantothenate and coenzyme A in bacterial growth. J. Bacteriol. 92:707-713. 1966.-The effect of environmental pantothenate levels on the growth of Streptococcus faecalis 9790 was studied in terms of growth rate, depletion phenomena, cellular coenzyme A (CoA) content, and differential rates of wall and...
Pantothenate kinase is an essential enzyme in the bacterial life cycle. It catalyzes the phosphorylation of pantothenate (vitamin B5) to 4'-phosphopantothenate, the first step in the coenzyme A biosynthetic pathway. The enzyme from Mycobacterium tuberculosis, MW 35.7 kDa, has been cloned, expressed, purified and crystallized in two different trigonal crystal forms, both belonging to space group...
Mutations in nuclear genes associated with defective coenzyme A biosynthesis have been identified as responsible for some forms of neurodegeneration with brain iron accumulation (NBIA), namely PKAN and CoPAN. PKAN are defined by mutations in PANK2, encoding the pantothenate kinase 2 enzyme, that account for about 50% of cases of NBIA, whereas mutations in CoA synthase COASY have been recently r...
The nutritional equivalence of pantothenate and p-aminobenzoate for the growth of Bacterium linens and the reversal of growth inhibition by sulfanilamide and salicylate has indicated a biochemical relationship between pantothenate and p-aminobenzoate (Purko et al., 1953). Recently, Pomper (1952) observed that the pantothenate, as well as the methionine, histidine, and adenine requirements of a ...
Hallervorden Spatz Disease is a rare neurodegenerative disorder with the prevalence of one to three per million. The onset of symptoms is usually in late childhood and early adolescence. However, some cases of the disease were reported in adulthood which could be familial or sporadic. The familial cases are autosomal-recessive resulting from mutation in the pantothenate kinase 2 gene located on...
