Total syntheses of 4-methylthio-3-butenylisothiocyanate (MTBI, 1) and raphanusanin (2), which are growth inhibitors involved in the phototropism radish (Raphanus sativus) hypocotyls, were achieved from commercially available thiolane four five steps, respectively. MTBI (1) was prepared via ring opening with sodium azide, double bond introduction by oxidation, isothiocyanation. Cyclization to (2...

The roots curl in naphthylphthalamic acid1 (rcn1) mutant of Arabidopsis (Arabidopsis thaliana) has altered auxin transport, gravitropism, and ethylene response, providing an opportunity to analyze the interplay between ethylene and auxin in control of seedling growth. Roots of rcn1 seedlings were previously shown to have altered auxin transport, growth, and gravitropism, while rcn1 hypocotyl el...

Actinomycin D inhibited the synthesis of poly(A)-containing messenger RNA in healthy soybean (Glycine max [L.] Merr. cv. Harosoy 63) hypocotyls and in hypocotyls inoculated with the pathogenic fungus Phytophthora megasperma var. sojae A. A. Hildb., but had little effect on protein synthesis within 6 hours. Blasticidin S, conversely, inhibited protein synthesis in the hypocotyls without exhibiti...

Etiolated hypocotyls of the resistant soybean (Glycine max [L.] Merr.) cultivar Harosoy 63 became susceptible to Phytophthora megasperma (Drechs.) f.sp. glycinea (Hildeb.) Kuan and Erwin race 1 after treatment with abscisic acid. Susceptibility was expressed by increases in lesion size and a major decrease in accumulation of the isoflavonoid phytoalexin, glyceollin. In untreated hypocotyls, act...

Glucosinolates are secondary metabolites derived from amino acids. Upon hydrolysis by myrosinases, they produce a variety of biologically active compounds. In this study, the glucosinolate-myrosinase system was characterized in Arabidopsis suspension cells. A total of seven glucosinolates were identified and the myrosinase activity was determined. Plant suspension cells have been used as model ...

Physiological changes were examined in the amount of a 50-kDa glycoprotein (gp50) that was recovered in a nuclear fraction from hypocotyls of mung bean (Vigna radiata) seedlings. Immunoblot analysis indicated that the glycoprotein was present in hypocotyls and epicotyls from 4- and 5-day-old seedlings but not in hypocotyls from 2-day-old seedlings. The glycoprotein was not detected in leaves or...

A cDNA clone corresponding to a gibberellin-responsive gene (CRG16) was isolated from cucumber hypocotyls. CRG16 was deduced to encode an extremely hydrophobic protein of 65 amino acids. The deduced sequence exhibited no significant homology to other proteins. Levels of CRG16 mRNA reflected the gibberellin-induced elongation of cucumber hypocotyls.

The effects of increasing NaCl concentrations on biomass, superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and phenylalanine ammonia-lyase (PAL) in Jatropha curcas L. seedlings were investigated. The fresh weights of cotyledons and radicles with increasing NaCl concentrations decreased progressively, and the fresh weight of hypocotyls reached the lowest level at NaCl concentration ...

Elongation of hypocotyls of sunflower can be promoted by gibberellins (GAs) and inhibited by ethylene. The role of these hormones in regulating elongation was investigated by measuring changes in both endogenous GAs and in the metabolism of exogenous [(3)H]- and [(2)H(2)]GA(20) in the hypocotyis of sunflower (Helianthus annuus L. cv Delgren 131) seedlings exposed to ethylene. The major biologic...

The actin cytoskeleton is hypothesized to play a major role in gravity perception and transduction mechanisms in roots of plants. To determine whether actin microfilaments (MFs) are involved in these processes in stem-like organs, we studied gravitropism in Arabidopsis inflorescence stems and hypocotyls. Localization studies using Alexa Fluor-phalloidin in conjugation with confocal microscopy d...
