چکیده مقاله:

Introduction: The choice of optimal radionuclides for radioimmunotherapy depends on several factors, especially the radionuclide and antibody. The dosimetric characteristics of a non-internalizing and an internalizing monoclonal antibody (MAb) labeled with beta emitting radionuclides were investigated. Methods: Using Geant4-DNA Monte Carlo simulation, we carry out dosimetric calculations for different subcellular distributions of beta-emitting radionuclides; 131I, 177Lu, 64Cu, 186Re and 153Sm. Results:The dependency of theradialdose profiles on the energy spectra of   electrons (beta particles and Auger and internal conversion electrons) and also their relative yield of emission is clear. The highest difference between the radionuclides tested was observed when the activity was localized in the nucleus. There was not considerable difference in the nucleus dose when radionuclides were localized in cytoplasm and over the cell membrane. Conclusion: There is a very significant increase in the dose deposited to the nucleus if 153Sm localized at the nucleus. Although subcellular localization of activity isn’t a critical factor for beta emitting radionuclides, but the use of internalizing MAbs leads to an increase in nucleus dose and to the killing of single cells in addition to the tumors.

Bismuth-212-labeled anti-Tac monoclonal antibody: alpha-particle-emitting radionuclides as modalities for radioimmunotherapy.

Anti-Tac, a monoclonal antibody directed to the human interleukin 2 (IL-2) receptor, has been successfully conjugated to the alpha-particle-emitting radionuclide bismuth-212 by use of a bifunctional ligand, the isobutylcarboxycarbonic anhydride of diethylenetriaminepentaacetic acid. The physical properties of 212Bi are appropriate for radioimmunotherapy in that it has a short half-life, deposit...

Pediatric radiation dosimetry for positron-emitting radionuclides using anthropomorphic phantoms.

PURPOSE Positron emission tomography (PET) plays an important role in the diagnosis, staging, treatment, and surveillance of clinically localized diseases. Combined PET/CT imaging exhibits significantly higher sensitivity, specificity, and accuracy than conventional imaging when it comes to detecting malignant tumors in children. However, the radiation dose from positron-emitting radionuclide t...

Assessment of the Specific Activity of Alpha- and Beta-emitting Radionuclides in Groundwater, Anka, Nigeria

Introduction: After the lead poisoning outbreak came to light in Zamfara State, Nigeria, people living near gold mines were worried about environmental safety, especially drinking water quality. This study examined the gross alpha and gross beta activity concentrations in groundwater in different locations of Anka, Zamfara State, Nigeria, to measure the possible radiation dose and potential hea...

New calculations for internal dosimetry of beta-emitting radiopharmaceuticals.

The calculation of absorbed dose from internally incorporated radionuclides is based on the so-called specific absorbed fractions (SAFs) which represent the fraction of energy emitted in a given source region that is absorbed per unit mass in a specific target organ. Until recently, photon SAFs were calculated using MIRD-type mathematical phantoms. For electrons, the energy released was assumed...

Evaluation of Factors Influencing Antibody Reduction for Development of Antibody Drug Conjugates

Background: Reduction/alkylation is one of the leading strategies for the development of antibody drug conjugates (ADCs). Precise control of the reduction process would not only yield a defined number of free thiols per antibody but also result in development of more homogenous conjugates. Methods: In the present study, we investigated the effect of various dithiothreitol (DTT) concentrations, ...
